Megjegyzések:Transglutaminase 2 (TG2) has been known for along time to be associated with the in vivo apoptosis programof various cell types, including T cells. Though theexpression of the enzyme is strongly induced in mousethymocytes following apoptosis induction in vivo, no significantinduction of TG2 can be detected, when thymocytesare induced to die by the same stimuli in vitro indicatingthat signals arriving from the tissue environment arerequired for the proper in vivo induction of the enzyme.Previous studies from our laboratory have demonstratedthat two of these signals, transforming growth factor-?(TGF-?) and retinoids, are produced by macrophagesengulfing apoptotic cells. However, in addition to TGF-? and retinoids, engulfing macrophages produce adenosineas well. Here, we show that in vitro adenosine, adenosine,and retinoic acid or adenosine, TGF-? and retinoicacids together can significantly enhance the TG2 mRNAexpression in dying thymocytes. The effect of adenosineis mediated via adenosine A2A receptors (A2ARs) andthe A2AR-triggered adenylate cyclase signaling pathway.In accordance, loss of A2ARs in A2AR null mice significantlyattenuates the in vivo induction of TG2 followingapoptosis induction in the thymus indicating that adenosineindeed contributes in vivo to the apoptosis-related appearanceof the enzyme. We also demonstrate that adenosineis produced extracellularly during engulfment of apoptoticthymocytes, partly from adenine nucleotides released viathymocyte pannexin-1 channels. Our data reveal a novelcrosstalk between macrophages and apoptotic cells, inwhich apoptotic cell uptake-related adenosine productioncontributes to the appearance of TG2 in the dyingthymocytes.
