Megjegyzések:The possibility that endocrine factors mayinfluence the clinical course of malignant melanoma issuggested by the superior survival data of women. In preclinicalmodels we observed a higher rate of colony formationby human melanoma cells in male compared tofemale SCID mice, but only in the case of the liver and notin other organs. The gender difference could be seen at anearly phase of colony formation. On the other hand, in ourhuman melanoma cell lines we failed to detect steroidreceptor protein expression, and treatment with sexhormones did not considerably influence their in vitrobehavior. Investigating the possible contribution of hostcells to the observed gender difference, we performedin vivo blocking experiments applying pretreatment of theanimals with Kupffer cell inhibitor gadolinium chlorideand the NK cell inhibitor anti-asialo GM1 antibody. WhileKupffer cell blockade enhanced melanoma liver colonizationequally in the two sexes, a more prominent increasewas observed in female than in male mice in the case ofNK cell inhibition. Further supporting the importance ofNK cells in the lower liver colonization efficiency ofmelanoma cells in females, gender difference in colonyformation was lost in NSG mice lacking NK activity.Although in humans no organ selectivity of gender differencein melanoma progression has been observedaccording to data in the literature, our results possiblyindicate a contribution of natural host defense mechanismsto gender difference in survival of patients with melanomaor other tumor types as well.
