Megjegyzések:The western Palaearctic water frogs in the genus Pelophylax comprise several distinct biological species and three hybridogenetic hybrid forms. In this study, we focus on the Pelophylax esculentus complex, which consists of two sexual species, Pelophylax ridibundus and Pelophylax lessonae and their hybridogenetic hybrid, Pelophylax esculentus. We used variation in serum albumin intron 1 (SAI?1) and 15 microsatellite loci for taxonomic and ploidy level identification of 164 individuals collected from three different types of wetland habitats in eastern Hungary at Hortobágy National Park (HNP). We detected the presence of all members of the P. esculentus complex in the studied localities. In one locality all three water frog taxa occurred syntopically, while in other localities P. ridibundus and P. esculentus dominated. The genomic composition of the 63 examined hybrid specimens analysed with microsatellite loci showed that only the diploid genotypes were presented in the studied populations. We used population genetics (allelic richness, gene diversity, multilocus genotypes and multilocus disequilibrium) to infer the breeding system of water frogs at HNP. Our data indicates that at least in two populations hybrids form gametes with clonally transmitted P. ridibundus genome and can only make new hybrid generation by mating with P. lessonae.

Megjegyzések:Chytridiomycosis, caused by the fungal pathogen Batrachochytrium dendrobatidis (Bd), is anemerging infectious disease implicated in declines of amphibian populations around theglobe. In Hungary the presence of the chytrid fungus from elevated habitats was documentedbefore but little is known about the distribution in the lowlands. The Western Palearcticwater frogs of the genus Pelophylax include a series of about 12 morphologically similar speciesin Europe. The Pelophylax species complex consists three species in Hungary: P. ridibundus,P. lessonae and the hybrid P. kl. esculentus. Our sampling area situated in the HortobágyNational Park and we collected 29 Pelophylax spp. from two localities associated with flowingwater and 30 specimens from the area of fish ponds and from there additionaly non-invasivesamples (19) becoming from skin swabbing. We used newly developed primers for distinguishingamong the water frog species, and real-time PCR for detecting the Bd zoospores.Based on our molecular taxonomy findings the species composition of the investigated populationsof water frogs consists the P. ridibundus and the hybrid P. kl. esculentus. Our resultsclearly showed that the population of water frogs connected with flowing water were infectedwith Bd, while the population inhabiting the fish ponds were uninfected. The prevalence was18.9% for the whole population together, while 51.7% for the frogs from the flowing water.Prevalence was 37.1% for P. ridibundus and 8.3% for P. kl esculentus. The mean intensity was10.13 GE (genome equivalent zoospore quantity).

Megjegyzések:Parasites are becoming recognized as significant participants in the evolutionary game and areseen as perfect model systems for general evolutionary studies. Our present knowledge of theadult platyhelminth parasites of vertebrates in Hungary especially in the Hortobágy NationalPark is defective. Our aim of examination was to reveal the helminth fauna of the Pelophylaxesculentus complex and compare our findings with the results of previous studies and comparethe faunistical changes now with a baseline from more than 40 years ago. Fifty-nineindividuals of Pelophylax spp. were collected between May and September of 2012 (35 P. ridibundusand 24 P. kl. esculentus) and we found the following species of parasites. Digeneans:Haematoloechus variegatus, Opisthioglyphe ranae, Diplodiscus subclavatus, Pleurogenoides medians;nematodes: Oswaldocruzia filiformis, Rhabdias esculentarum; acanthocephalans: Acanthocephalusranae. Rhabdias esculantarum is a new species for the Hungarian fauna and P. ridibundusrepresents a new host record for Rh. esculentarum in Europe, while D. subclavatus isalso a new species for the fauna of the Hortobágy. The prevalence was 84.7% [95% CI:73,05?92,05] in the examined water frog population. The mean intensity was 2.44 (? 1.49)helminth species per host. No host contained more than 6 species: 15 (25.4%) of 59 Pelophylaxspp. harbored 1 species, 13 (22%) harbored 2 species, 12 (20%) harbored 3 species, 6 (10%)harbored 4 species, 3 (5%) harbored 5 species, and 1 (1.6%) harbored 6 species.

Megjegyzések:Az elmúlt évtizedek során egyre inkább előtérbe került azon felismerés, hogy a parazita szervezetek a biológiai sokféleség szignifikáns komponenseit képezik, továbbá kiváló modell rendszerekként viselkednek általános evolúcióbiológiai és ökológiai kutatásokban.Vizsgálatunk alapját a Hortobágyi Nemzeti Park területén élő zöld békákon végzett korábbi tanulmányok adták, amelyek publikálása óta, az elmúlt 30 évben nem történtek hasonló fel-mérések a területen. Három vizes élőhelyről (Egyek-Pusztakócs-mocsárrendszer, Nádudvar-csatornák, Hortobágy-halastavak) gyűjtöttük Pelophylax sp. egyedeket 2012 májusa és 2013 augusztusa között. A zöld békák meghatározása molekuláris taxonómiai módszerekkel történt.A következő parazita taxonokat találtuk. Digenea: Haematoloechus variegates, Opisthio-glyphe ranae, Diplodiscus subclavatus, Pleurogenes claviger, Pleurogenoides medians, Nematoda: Oswaldocruzia filiformis, Rhabdias esculentarum, Acanthocephala: Acantho-cephalus ranae. A R. esculentarum új faj a magyar faunára tekintve, ahogy a Pelophylax ridibundus is új gazda rekord a faj számára Európában. A D. subclavatus és a P. claviger és a P. medians szintén új faj a hortobágyi faunára nézve. A prevalencia 76.2 % [95%CI:66.89-83.78] a vizsgált zöldbéka populációban. A féreg taxonok átlagos intenzitása 2.44 [95%CI:2.17-2.71] parazita faj gazdánként. Az eredményeink szignifikáns különbséget mutattak a parazita fauna összetételében a korábbi vizsgálatokhoz képest.

METHODS: We collected 101 individuals of the Pelophylax esculentus complex (Ranidae) in 2012 and 2013 from sites associated with flowing water, a fish pond system and a wetland marsh system. RESULTS: We found the following species: Digeneans: Haematoloechus variegatus, Opisthioglyphe ranae, Diplodiscus subclavatus, Pleurogenes claviger, Pleurogenoidesmedians Nematodes: Oswaldocruzia filiformis, Rhabdias esculentarum. Acanthocephala: Acanthocephalus ranae. R. esculantarum is a new species for the Hungarian fauna and P.ridibundus represents a new host record for R. esculentarum, while D. subclavatus, P.claviger and P. medians is also a new species for the helminthofauna of the Hortobágy National Park. The overall prevalence was 76.2 % [95%CI:66.89-83.78] in the examined water frog population. The overall mean intensity was 2.44 [95%CI:2.17-2.71] helminth species per host. CONCLUSIONS: Our current findings showed significant discrepancy from the results of baseline inventories carried out 30 years ago in HNP, but the reasons are not clear. We suspect that the helminth species previously reported that we did not encounter are restricted to a host we have not yet collected.

Megjegyzések:The western palearctic water frogs in the genus Pelophylax include twelve species with partially overlapping morphology. Some of these species can form hybridogenetic complexes. Such type is the Pelophylax esculentus complex formed by two sexual species: Pelophylax ridibundus (genotype RR), Pelophylax lessonae (genotype LL), and their hybridogenetic hybrid Pelophylax esculentus (genotype LR, LRR or LLR). This study was conducted in the Hortobágy National Park which is the largest contiguous alkaline steppe in Europe covering 80.000 hectares. Altogether 164 water frogs were captured during the years 2012-2014 from three different types of wetland habitats. Due to the high rate of hybridization and partially overlapping morphological features among water frogs we used molecular taxonomic techniques to distinguish between the parental species and the hybrid. This method is based on the PCR products size differences from the amplification of the serum albumin intron-1 (SAI-1) gene fragment. We also used 15 microsatellite loci to distinguish between the three different genotypes in hybrid individuals. Based on fragmentary analysis we detected the presence of all the members of P. esculentus complex. From 164 samples we determined the taxonomic status of 101 P. ridibundus, 1 P. lessonae and 62 P. esculentus individuals. Based on microsatellite analysis we found that only the diploid (LR) genotype was presented in hybrid individuals

Megjegyzések:Pelophylax génuszba tartozó és Nyugat-palearktiszi elterjedéssel bíró zöldbékák közé jelenleg 12 fajt sorolunk, amelyek egy részénél megfigyelhető hibridogenetikus fajkomplexek jelenléte. Vizsgálatunkban a Magyarországon is előforduló kecskebéka fajkomplexre (Pelophylax esculentus komplex) irányítottuk figyelmünket. Ennek a komplexnek a tagjai a tavi béka (Pelophylax ridibundus, genotípus RR) a kis tavibéka (Pelophylax lessonae, genotípus LL) illetve ezen két szülői faj hibridizációjából létrejövő kecskebéka (Pelophylax esculentus, genotípus LR). A hibrid egyedek a szülői fajoktól morfológiai jellegeik alapján nehezen elkülöníthetőek, ezért molekuláris taxonómiai megközelítést (polimorfia detektálása a szérumalbuminintron 1 génben) használtunk arra, hogy az egyes élőhelyeken a fajösszetételt meghatározzuk, továbbá a hibrid egyedek genotípusát mikroszatellita vizsgálattal feltárjuk, majd populáció genetikai indexek számításával következtessünk a szaporodási rendszerükre. A három éves mintagyűjtés során 164 zöldbékától vettünk DNS mintát a Hortobágyi Nemzeti Park (HNP) három különböző élőhelyi paraméterekkel rendelkező vizes élőhelyén. A komplex mind a három tagjának jelenlétét észleltük a mintavételi területen. Egyik vizes élőhelyen mindhárom taxon együtt fordult elő, míg a másik két élőhelyen a tavi béka és a kecskebéka együttes előfordulását detektáltuk. A ploiditás vizsgálat során 63 hibrid kecskebéka genotípusát analizáltuk és azt találtuk, hogy az egyedek diploid (LR) genotípussal rendelkeztek. Triploid genotípusú (LLR és LRR) békákat nem találtunk. Az eredményeink azt indikálják, hogy a három élőhelyből legalább kettőben LE (lessonae-esculenta) szaporodási rendszert figyelhetünk meg, ahol a hibridek a ridibundus (R) genomjaikat klonális módon adják tovább a következő hibrid generációnak és csak a kis tavi békával visszakereszteződve képes sikeresen szaporodni.

Megjegyzések:A parazitizmus a populációk közötti kölcsönhatások egyik alapvető formája, elég, ha csak arragondolunk, hogy a Földön élő növények és állatok 40-50%-a parazita, vagy patogén életciklusavalamely szakaszában. Jelenlegi tudásunk a hazai gerincesek, jelen esetben a kétéltűek endoparazitaféreg (Trematoda, Nematoda, Acanthocephala) taxonjairól hiányos. Mintaterületünket a HortobágyiNemzeti Park északi részén fekvő Halastavak tórendszer és a kissé délre található csatornák ésöntöző vizek alkotják. Korábban a Nemzeti Park területéről Edelényi (1974) illetve Murai ésmunkatársai (1983) gyűjtöttek parazita mintákat kétéltűekből. 2012 májusa és szeptembere közöttkézzel és merítőhálóval begyűjtött, illetve az út mentén elütött példányokból származó ?törzsanyag"59 Pelophylax spp.-t számlált. Az állatok begyűjtése mellett DNS mintát is izoláltunk a hátsó lábegyik ujjpercéből. Egy újonnan kifejlesztett gyors és költséghatékony PCR metódus és agarózgélelektroforézis segítségével, sikerült fajra beazonosítani a P. esculentus komplex tagjait, amely amagas hibridizációs ráta miatt morfológia alapján nehézkes lett volna. Ez az eredmény az elsőmolekuláris taxonómia vizsgálat a hortobágyi, sőt hazai zöld béka populációkra vonatkozóan. Azeredmények alapján elmondható, hogy tavi béka (P. ridibundus) és a hibrid kecskebéka (P. kl.esculentus) alkotja a terület zöldbéka populációját. Az állatok éterrel történő túlaltatása után azemésztő traktust elválasztottuk a többi szervtől, majd azokat külön vizsgáltuk, mint egyesmikrohabitatokat a paraziták szempontjából. A gazdák fajszintű ismerete alapján meghatározhatóazok parazita terhelése. 59 egyedből 50 bizonyult legalább egy parazita fajjal fertőzöttnek, ami 84,7%-os prevalenciát mutat. A korábban leírt parazita fajok közül nem találkoztunk a Haplometracylindracea, Haematoloeuchus asper, Cephalogonimus retesus, Clinostomum complanatum,Cosmocerca ornata és Gorgodera cygnoides egyedeivel. Viszont a Diplodiscus subclavatus és aRhabdias spp. új faj a hortobágyi faunára. Továbbá a Murai és mts. (1983) által Pleurogenes loosiinakhatározott közvetett fejlődésű métely a mintáink alapján inkább Pleurogenoides medians-nakbizonyult.

Megjegyzések:The Document, Assess, Monitoring, Act (DAMA) protocol details an approach to integrating informationabout parasites into large-scale studies of biodiversity, climate change, and emerging diseases. This study represents an effort toput the DAMA protocol into practice. We collected 101 individuals of protected ranid frogs belonging to the Pelophylaxesculentus complex during 2012 and 2013 in the Hortoba?gy National Park (HNP) in eastern Hungary in an area where aninventory of amphibian helminths had been conducted 40 yr previously. Collecting sites included flowing water, a fish pondsystem, and a wetland marsh system. We found the following helminth species: Digeneans: Diplodiscus subclavatus,Haematoloechus variegatus, Opisthioglyphe ranae, Pleurogenes claviger, Pleurogenoides medians; Nematodes: Oswaldocruziafiliformis, Rhabdias esculentarum; and Acanthocephala: Acanthocephalus ranae. Rhabdais esculentarum is a newspecies for the Hungarian fauna and P. ridibundus represents a new host record for R. esculentarum while D. subclavatus,P. claviger, and P. medians are new species for the helminthofauna of the HNP. Our findings showed a significant discrepancyfrom the results of baseline inventories carried out 40 yr ago, although the reasons for this discrepancy are not clear. We suspectthat the previously reported helminth species that we did not encounter are restricted to Pelophylax lessonae, a host we have notyet collected at this location, but factors associated with climate change or anthropogenic impacts cannot be ruled out.

Megjegyzések:Batrachochytrium dendrobatidis (Bd) is a fungal pathogen which causes the emerging infectious disease chytridiomycosis. Bd presents low host specificity and threatens amphibians worldwide, thus systematic inventory is the key in order to detect and mitigate the effects of the disease. Extensive data collection was conducted in Hungary in 2009-2015 from 14 different areas. Combined data ? recent field sampling on 16 taxa and the examination of archived Bombina spp. specimens ? from 1360 individuals were analysed with qPCR. Two sentinel taxa, Bombina variegata and the members of the Pelophylax esculentus complex were marked to monitor the occurrence of Bd in two core areas (Bakony Mts) and Hortobágy National Park, respectively) of sampling. Climatic variables were also examined in core areas to test their effect on prevalence and infection intensity. Among the sixteen sampled amphibian taxa seven tested positive for Bd and the overall prevalence in Hungary was 7.46%. Among the ethanol-fixed Bombina spp. individuals Bd was not detected. In the first core area (Bakony Mts) the overall prevalence in B. variegata was 10.32% and juvenile individuals showed significantly higher prevalence than adults. On the other hand there was a significant negative relationship between infection prevalence and monthly mean air temperature. Finally, in the other core area (Hortobágy National Park) the overall prevalence in P. esculentus complex was 13.00%, and no differences were found in prevalence or infection intensity between sexes, sampling years or age classes.
