Megjegyzések:PAF and related antifungal proteins are promising antimicrobial agents. They have highly stable folds around room temperature due to the presence of 3-4 disulfide bonds. However, unfolded states persist and contribute to the thermal equilibrium in aqueous solution, and low-populated states might influence their biological impact. To explore such equilibria during dimethyl sulfoxide (DMSO)-induced chemical unfolding, we studied PAF and its inactive variant PAFD19S using nuclear magnetic resonance (NMR) and differential scanning calorimetry (DSC). According to the NMR monitoring at 310 K, the folded structures disappear above 80 v/v% DMSO concentration, while the unfolding is completely reversible. Evaluation of a few resolved peaks from viscosity-compensated 15N-1H HSQC spectra of PAF yielded G = 23 ± 7 kJ/M as the average value for NMR unfolding enthalpy. The NMR-based structures of PAF and the mutant in 50 v/v% DMSO/H2O mixtures were more similar in the mixed solvents then they were in water. The 15N NMR relaxation dynamics in the same mixtures verified the rigid backbones of the NMR-visible fractions of the proteins; still, enhanced dynamics around the termini and some loops were observed. DSC monitoring of the Tm melting point showed parabolic dependence on the DMSO molar fraction and suggested that PAF is more stable than the inactive PAFD19S. The DSC experiments were irreversible due to the applied broad temperature range, but still suggestive of the endothermic unfolding of PAF.

Megjegyzések:Their emerging nature as multifunctional effectors explains the large interest to monitor glycan binding to galectins and to define bound-state conformer(s) of their ligands in solution. Basically, NMR spectroscopy facilitates respective experiments. Towards developing new and even better approaches for these purposes, extending the range of exploitable isotopes beyond 1H, 13C, and 15N offers promising perspectives. Having therefore prepared selenodigalactoside and revealed its bioactivity as galectin ligand, monitoring of its binding by 77Se NMR spectroscopy at a practical level becomes possible by setting up a 2D 1H, 77Se CPMG-HSQBMC experiment including CPMG-INEPT long-range transfer. This first step into applying 77Se as sensor for galectin binding substantiates its potential for screening relative to inhibitory potencies in compound mixtures and for achieving sophisticated epitope mapping. The documented strategic combination of synthetic carbohydrate chemistry and NMR spectroscopy prompts to envision to work with isotopically pure 77Se-containing ?-galactosides and to build on the gained experience with 77Se by adding 19F as second sensor in doubly labeled glycosides.

Megjegyzések:Human galectin-3 (hGal-3) is involved in a variety of biological processes and is implicated in wide range of diseases. As a result, targeting hGal-3 for clinical applications has become an intense area of research. As a step towards the development of novel hGal-3 inhibitors, we describe a study of the binding of two Se-containing hGal-3 inhibitors, specifically that of di(?-Dgalactopyranosyl)selenide (SeDG), in which two galactose rings are linked by one Se atom and a di(?-D-galactopyranosyl)diselenide (DSeDG) analogue with a diseleno bond between the two sugar units. The binding affinities of these derivatives to hGal-3 were determined by 15N-1H HSQC NMR spectroscopy and fluorescence anisotropy titrations in solution, indicating a slight decrease in the strength of interaction for SeDG compared to thiodigalactoside (TDG), a well-known inhibitor of hGal-3, while DSeDG displayed a much weaker interaction strength. NMR and FA measurements showed that both seleno derivatives bind to the canonical S face site of hGal-3 and stack against the conserved W181 residue also confirmed by X-ray crystallography, revealing canonical properties of the interaction. The interaction with DSeDG revealed two distinct binding modes in the crystal structure which are in fast exchange on the NMR time scale in solution, explaining a weaker interaction with hGal-3 than SeDG. Using molecular dynamics simulations, we have found that energetic contributions to the binding enthalpies mainly differ in the electrostatic interactions and in polar solvation terms and are responsible for weaker binding of DSeDG compared to SeDG. Selenium-containing carbohydrate inhibitors of hGal-3 showing canonical binding modes offer the potential of becoming novel hydrolytically stable scaffolds for a new class of hGal-3 inhibitors.

Megjegyzések:Several ellagitannins inhibited the activity of protein phosphatase-1 (PP1) and -2 A (PP2A) catalytic subu-nits (PP1c and PP2Ac) with preferential suppression of PP1c over PP2Ac. The inhibitory potency for PP1cfollowed the order of tellimagrandin I>mahtabin A>praecoxin B>1.2-Di-O-galloyl-4.6-(S)-HHDP-b-D-glu-copyranose>pedunculagin with IC50values ranging from 0.20mM to 2.47mM. The interaction of PP1c andtellimagrandin I was assessed by NMR saturation transfer difference, surface plasmon resonance, isother-mal titration calorimetry, and microscale thermophoresis based binding techniques. Tellimagrandin I sup-pressed viability and phosphatase activity of HeLa cells, while mahtabin A was without effect. Conversely,mahtabin A increased the phosphorylation level of SNAP-25Thr138and suppressed exocytosis of corticalsynaptosomes, whereas tellimagrandin I was without influence. Our results establish ellagitannins as par-tially selective inhibitors of PP1 and indicate that these polyphenols may act distinctly in cellular systemsdepending on their membrane permeability and/or their actions on cell membranes.

Megjegyzések:A new method is proposed that allows broadband homonuclear decoupled CLIP/CLAP-HSQC NMR spectra to be acquired at virtually no extra cost in measurement time. The real-time (windowed) acquisition protocol applied follows a scheme recently devised for recording pure shift (broadband homonuclear decoupled) heteronuclear single quantum correlation (HSQC) spectra. To minimize systematic errors in the apparent coupling constants obtained using real-time homonuclear decoupling, we extended the acquisition scheme to include cycling of radiofrequency pulse phases both from chunk to chunk during windowed acquisition, and from scan to scan during time averaging, allowing robust coupling constant measurement. The new real-time pure shift CLIP/CLAP-HSQC experiments are designed to speed up coupling constant determination, to increase the sensitivity of measurement in favorable cases, and to simplify the extraction of accurate one-bond heteronuclear couplings from pure in- or anti-phase doublets using automatic peak picking. The scope and limitations of the method are discussed, and a variety of experimental tests are reported.
