Megjegyzések:The in situ control of redox insult in human organs is of major clinical relevance, yet remains incompletely understood. Activation of nuclear factor (erythroid-derived 2)-like 2 (Nrf2), the "master regulator" of genes controlling cellular redox homeostasis, is advocated as a therapeutic strategy for diseases with severely impaired redox balance. It remains to be shown whether this strategy is effective in human organs, rather than only in isolated human cell types. We have therefore explored the role of Nrf2 in a uniquely accessible human (mini-) organ: scalp hair follicles. Microarray and qRT-PCR analysis of human hair follicles after Nrf2 activation using sulforaphane identified the modulation of phase II metabolism, reactive oxygen species clearance, the pentose phosphate pathway, and glutathione homeostasis. Nrf2 knockdown (small interfering RNA) in cultured human hair follicles confirmed the regulation of key Nrf2 target genes (i.e., heme oxygenase-1, NAD(P)H dehydrogenase, quinone 1, glutathione reductase, glutamate-cysteine ligase catalytic subunit, ABCC1, peroxiredoxin 1). Importantly, Nrf2 activation significantly reduced reactive oxygen species levels and associated lipid peroxidation. Nrf2 preactivation reduced premature catagen and hair growth inhibition induced by oxidative stress (H2O2 or menadione), significantly ameliorated the H2O2-dependent increase in matrix keratinocyte apoptosis and reversed the reactive oxygen species-induced reduction in hair matrix proliferation. This study thus provides direct evidence for the crucial role of Nrf2 in protecting human organ function (i.e., scalp hair follicles) against redox insult.

Megjegyzések:Rosacea is a common, chronic inflammation of sebaceous gland-rich facial skin characterized by severe skin dryness, elevated pH, transepidermal water loss, and decreased hydration levels. Until now, there has been no thorough molecular analysis of permeability barrier alterations in the skin of rosacea patients. Thus, we aimed to investigate the barrier alterations in papulopustular rosacea (PPR) samples compared to healthy sebaceous gland-rich (SGR) skin, using RNASeq analysis (n=8). Pathway analyses by Cytoscape ClueGo revealed 15 significantly enriched pathways related to skin barrier formation. RT-PCR and immunohistochemistry were used to validate the pathway analyses. The results showed significant alterations in barrier components in PPR samples compared to SGR, including the cornified envelope and intercellular lipid lamellae formation, desmosome and tight junction organizations, barrier alarmins, and antimicrobial peptides. Moreover, the barrier damage in PPR was unexpectedly similar to atopic dermatitis (AD); this similarity was confirmed by immunofluorescent staining. In summary, besides the well-known dysregulation of immunological, vascular, and neurological functions, we demonstrated prominent permeability barrier alterations in PPR at the molecular level, which highlight the importance of barrier repair therapies for rosacea.

Megjegyzések:We have previously shown that the endocannabinoid anandamide and the phytocannabinoid ( )-?9-trans-tetrahydrocannabinol reduce hair shaft elongation via promoting catagen in a CB1 receptor-dependent manner. By using microdissected human scalp hair follicles (HFs), as well as cultured primary human outer root sheath keratinocytes (ORSKs), now we assessed the effects of the non-psychotropic phytocannabinoid ( )-cannabidiol (CBD), which does not activate CB1, but exerts complex anti-acne effects in sebocytes, and is currently being investigated in a phase II clinical trial in moderate to severe acne. We found that 0.1 ?M CBD promoted hair shaft production, and suppressed Toll-like receptor 3 activation-induced pro-inflammatory response of cultured ORSKs in an adenosine receptor-dependent manner, whereas at 10 ?M it suppressed HF growth, which was accompanied by the induction of premature catagen entry. Importantly, potent pro-catagen concentrations of CBD induced Ca2+-influx to ORSKs via the mostly Ca2+-permeable TRPV4 ion channels, activation of which was recently demonstrated to induce premature catagen entry (Szabó et al. JID, 2018). Thus, data of this pilot, exploratory study invite the hypothesis that low, adenosine receptor-activating doses of CBD could become useful as adjunct therapy for inflammatory hair loss disorders, whereas high-dose, TRPV4-activating CBD is an attractive candidate inhibitor of unwanted hair growth.
