Megjegyzések:The distribution of the Imperial Eagle (Aquila heliaca) in the Carpathian Basin is not continuous, since western andeastern breeding pairs are separated by 150 km from each other in Slovakia, and 70 km in Hungary. In the present study our aimwas to examine whether this geographical distance has resulted in any genetic separation between the Western and Eastern Slovakbreeding groups. We have used 132 shed feathers and 128 blood samples collected in the fields geographically representingthe whole of the Slovak breeding population, and included all juveniles ringed between 2004 and 2006. After successful DNAextractions we have determined the sex, microsatellite DNA-profiles and mtDNA control region haplotypes of the specimens.Data were integrated in a common Hungarian-Slovak "DNA-fingerprint" database, making identification of the same specimenpossible when recaptured. Based on a subsample of the collected individuals, the genetic structure of the Slovak population wastested using ten microsatellite loci and mtDNA control region haplotypes, and marginally significant genetic differentiation wasfound between western and eastern subpopulations. These results suggest that, in spite of the large dispersal capacity of the species,a relatively small geographic distance can also decrease the exchange rate of individuals between subpopulations. As thisresult involves only samples from the northern part of the breeding area, major conclusions concerning genetic structure and gene flow of Imperial Eagles in the entire Carpathian Basin population cannot be drawn without sampling and analysing the southernsubpopulations in Hungary.

Megjegyzések:In this study we estimated the annual turnover rates, and so indirectly the possible maximum mortality rates of female Eastern Imperial Eagles (Aquila heliaca) in an expanding population. DNA was extracted from shed feathers collected in territories where at least three consecutive years of breeding occurred. As DNA samples could not be obtained at every studied nest in each year, minimum and maximum turnover rates were estimated. The calculated rates, 27.7% (minimum) and 35.5% (maximum), are much higher than what could be expected based on studies of other raptor species. According to previous studies territory switching can occur frequently in an expanding population. However, we found evidence of it in only two of the 46 studied territories, thus we assume that despite the ongoing saturation some level of site fidelity is present in the population. Our findings suggest the high prevalence of mortality, most likely from poisoning and electrocution, but also predict a large number of floater individuals that fill up the vacant territories.

Megjegyzések:The development of genetic methods broadens the scope of non-invasive sample types. Large shed feathers are good material for genetic analyses, since they are easy to collect and a single feather can provide a sufficient amount of DNA for PCR-based methods. Previous studies have demonstrated that feather quality and type affect the DNA quality extracted from the feather tips. Besides the tip, the superior umbilicus part of the shaft is also proposed as an appropriate source of DNA. In our study, we examined whether some feather parameters (physical condition, type and size) and storage time affect amplification success of DNA extracted from the superior umbilicus of shed Eastern Imperial Eagle (Aquila heliaca) feathers. We also tested the effects of sunlight, temperature and humidity on DNA extracted from Domestic Goose (Anser anser domesticus) feathers with amplification of fragments of various sizes, modelling the environmental conditions of the moulting season. While good quality feathers usually provided sufficient DNA, the usability of the DNA extracted from moderate quality feathers were affected by feather type. DNA quality was influenced in order of importance by humidity, direct sunlight and heat. Our findings support the usability of DNA samples derived from the superior umbilicus of shed feathers, and help to schedule field work by careful consideration of our results about feather quality and environmental factors.
