Cím:Survey on antibiotic resistant bacteria in commercial fish ponds / Elshafia Ali Hamid Mohammed; Renáta Bőkényné Tóth; Gábor Kardos; Csongor Freytag; Károly Pál
Dátum:2024
Megjegyzések:Fish is a valuable source of proteins and commercial fish farms help to satisfy the demand. Hungarian farms produce a large amount of fresh fish every year, in many lakes countrywide. Due to the fish farming activities (the use of antibiotics for curing fish with bacterial infections) and the migrating birds (reservoir of pathogenic microbes), these lakes may be sources of pathogenic bacteria that confer antibiotic resistance. These bacteria may pose a threat not only to the fish but employees and consumers, as well. A joint project of the Institute of Metagenomics and the microbiology laboratory of the Institute of Food Sciences is to perform regular surveys on the microbiological quality of fish farm lake water, with emphasis on the detection of possible AMR bacteria, in accordance with the Farm to Fork Strategy. The survey started in 2023 and 10-10 lakes were sampled in two fish farms, respectively. After filtration of water samples, bacteria were isolated on vancomycine and Eosine Methylene Blue agar plates, then identified by MALDI-TOF. Though, numerous bacterial species were found, no traces of vancomycine resistant Enterococcus isolates were found yet.

Cím:Identification of Antibiotic-Resistant Bacteria in Commercial Fish Farms in Hungary / Elshafia Ali Hamid Mohammed; Renáta Bőkényné Tóth; Gábor Kardos; Pál Károly
Dátum:2024
Megjegyzések:The common carp (Cyprinus carpio) is a popular species in fish farms in Hungary (Europe). Since Hungary is a member state of the European Union, the use of antibiotics for growth promotion of animals has been banned since 2011. An even stricter legislation came into force in 2022 to inhibit the spread of antibiotic resistant bacteria in animals and humans, in accordance with the Farm to Fork Strategy. Nevertheless, new strains of AMR conferring bacteria may appear in fish farm lakes as a result of antibiotic treatment of bacterial infections, or due to wild birds as reservoirs. Antibiotic resistance genes (especially in form of mobile genetic elements, e.g. plasmids) may pose a serious threat to the fish production, health of employees, and environment. The current study aimed to survey the occurrence of antibiotic resistant bacteria and confirm their identity in water samples of commercial fish farms in Hungary. Twenty water samples were collected from two fish farms and subjected to bacterial isolation using a membrane filtration technique (pore size: 0.45-micrometer). Then the membranes were placed on different types of selected media (VRE, Eosin Methylene Blue and Endo's Medium). Bacterial colonies were subjected to matrix-assisted laser desorption ionization?timeof- flight mass spectrometry (MALDI-TOF MS) technique for species level identification. The following species were detected: Acinetobacter baumannii, A. calcoacetius, A. junii, A. pittii, A. tandoii, Bacillus pumilus, Comamonas aquatica, Enterococcus casseliflavus, E. gallinarum, Escherichia coli, Lactobacillus plantarum, Ochrobactrum intermedium, Pediococcus pentosaceus, Pseudomonas aeruginosa, P. alcaligenes, P. mendocina, P. otidis, P. putida and Stenotrophomonas maltophilia. In the next step, we will use antibiotic tests to investigate, whether these isolates have some unusual extrinsic antibiotic resistance. In addition, we will perform PCR experiments to detect the presence of virulence factors and toxin producing genes.

Cím:Effect of selected feed additives on growth performance and the intestinal microbiota of the Common carp (Cyprinus carpio) / Elshafia Ali Hamid Mohammed, Milán Fehér, Péter Bársony, Abdelhakam Esmaeil Mohamed Ahmed, Ayaz Mukarram Shaikh, Károly Pál
Dátum:2023
Megjegyzések:The research aimed to examine the impact of various feed additives on the growth performance and intestinal microbiota of the common carp (Cyprinus carpio). Three different feed additives including Bactocell (live lactic acid bacterium Pediococcus acidilactici CNCM I-4622 MA 18/5M), Levucell (probiotic derived from the fungus Saccharomyces cerevisiae boulardii CNCM I-1079) and YANG prebiotic (Saccharomyces cerevisiae AQP1260, S. cerevisiae AQP 12988 and Cyberlindnera jandinii AQP 12549) were tested at the concentration of 1% (10 g/1 kg of basic diets) in addition to the control (basic feed only). Fish with average body weight (g) of 932?163 were fed with supplemented diet for 45 days in a recirculation system (RAS). The RAS contains 12 units (size=1000 liter each unit, 8 fish per unit). Water temperature, pH, total dissolved solids, dissolved oxygen, NO-2 , NO-3 , and NH+4 concentrations were maintained within optimal levels. After the end of the feeding experiment, fish feces samples were collected and the bacterial community structure found in the intestines of common carp was analyzed through a metagenomic approach using 16S rDNA sequencing. The results indicated that there was no significant effect (P<0.05) of the tested feed additives (Bactocell, Levucell and YANG) on growth performance (final body weight), feed conversion ratio (FCR) and specific growth rate (SGR). The results of the metagenomic data of carp fed with a diet supplemented with YANG showed 11 different groups of bacterial phyla detected with the top three phyla, namely Fusobacteria (55%), Firmicutes (36%), and Proteobacteria (6%). In contrast, fecal samples from carp fed with 1% Bactocell recorded 10 phyla. The top three phyla were Proteobacteria (70%), Fusobacteria (21%), and Firmicutes (5%). A sample of carp fed with Levucell recorded 9 phyla, the top three phyla were Fusobacteria (50%), Proteobacteria (35%), and Firmicutes (14%). Finally, samples from carp fed with the basal diet (the control) recorded 14 phyla. The top three phyla recorded were Fusobacteriota (68%), Proteobacteria (27%), and Bacteroidota (3%). The study found that the tested feed additives did not significantly affect the growth performance of common carp. However, they did lead to variations in the composition of the intestinal microbiota of the common carp.
