Megjegyzések:UVB irradiation induces harmful photochemical reactions, including formation of Cyclobutane Pyrimidine Dimers (CPDs) in DNA. Accumulation of unrepaired CPD lesions causes inflammation, premature ageing and skin cancer. Photolyases are DNA repair enzymes that can rapidly restore DNA integrity in a light-dependent process called photoreactivation, but these enzymes are absent in humans. Here, we present a novel mRNA-based gene therapy method that directs synthesis of a marsupial, Potorous tridactylus, CPD-photolyase in cultured human keratinocytes. Pseudouridine was incorporated during in vitro transcription to make the mRNA non-immunogenic and highly translatable. Keratinocytes transfected with lipofectamine-complexed mRNA expressed photolyase in the nuclei for at least 2days. Exposing photolyase mRNA-transfected cells to UVB irradiation resulted in significantly less CPD in those cells that were also treated with photoreactivating light, which is required for photolyase activity. The functional photolyase also diminished other UVB-mediated effects, including induction of IL-6 and inhibition of cell proliferation. These results demonstrate that pseudouridine-containing photolyase mRNA is a powerful tool to repair UVB-induced DNA lesions. The pseudouridine-modified mRNA approach has a strong potential to discern cellular effects of CPD in UV-related cell biological studies. The mRNA-based transient expression of proteins offers a number of opportunities for future application in medicine.

Megjegyzések:Zinc is an essential microelement; its importance to the skin is highlighted by the severe skin symptoms in hereditary or acquired zinc deficiency, by the improvement of several skin conditions using systemic or topical zinc preparations and by the induced intracellular zinc release upon UVB exposure, which is the main harmful environmental factor to the skin. Understanding the molecular background of the role of zinc in skin may help gain insight into the pathology of skin disorders and provide evidence for the therapeutic usefulness of zinc supplementation. Herein, we studied the effects of zinc chloride (ZnCl2) exposure on the function of HaCaT keratinocytes, and the results showed that a non-toxic elevation in the concentration of extracellular zinc (100 mM) facilitated cell proliferation and induced significant alterations in the mRNA expression of NOTCH1, IL8, and cyclooxygenase-2. In addition, increased heme oxygenase-1 (HMOX1) expression and non-toxic generation of superoxide were detected in the first 4 h. Regarding the effects on the UVB-induced toxicity, although the level of cyclobutane pyrimidine dimers in the keratinocytes pre-treated with zinc for 24 h was reduced 3 h after UVB irradiation, significantly enhanced superoxide generation was observed 10 h after UVB exposure in the zinc pre-exposed cells. The overall survival was unaffected; however, there was a decrease in the percentage of early apoptotic cells and an increase in the percentage of late apoptotic plus necrotic cells. These results suggest that the exposure of human keratinocytes to non-toxic concentrations of ZnCl2 impacts gene expression, cell proliferation and the responses to environmental stress in the skin. It would be important to further examine the role of zinc in skin and further clarify whether this issue can affect our thinking regarding the pathogenesis of skin diseases.

Megjegyzések:Our aim was to assess whether the presence of highly active effector T-cells in atopic dermatitis (AD) is associated with changes in the number and/or function of regulatory T-cells (Tregs). Flow cytometry was utilised to determine the percentage of CD4+CD25brightCD127-/lowFOXP3+ and skin-homing CLA+CD4+CD25brightFOXP3+ Tregs in healthy controls and AD patients. The correlation between disease severity and Treg percentages was estimated. Treg suppressor activity and cell proliferation were measured after T-cell stimulation. Significantly increased percentages of Tregs were found in AD patients compared to healthy individuals, and significant correlation between the frequency of Tregs and disease severity was also detected. The otherwise normal suppressor activity of Tregs decreased in the presence of Staphylococcus enterotoxin B (SEB). In conclusion, the continuous presence of SEB can trigger an acquired functional impairment of Tregs in AD patients and the correlation between the increased frequency of Tregs and disease severity supports their important role in AD pathogenesis.

Megjegyzések:Atopic dermatitis (AD) is a chronic infl ammatory skin disease resulted partly of type I andtype IV hypersensitivity reactions triggered by T helper 2 (Th2) cells. The explanations behindthe Th2 dominance in the disease are still controversial. We addressed the question whetherthe Th2 cell domination is associated with a reduction and/or functional impairment ofCD4+CD25+FOXP3+ regulatory T cells (Treg) essential for the maintenance of Th1- Th2balance. Peripheral blood of AD patients with severe clinical symptoms and extremely highserum IgE levels (>2000 U/ml) were investigated. Flow cytometry was utilised to determinethe percentage of CD4+CD25brightFOXP3+ Tregs and CLA+CD4+CD25brightFOXP3+ Tregs in thesamples of AD patients and healthy controls. For detection of suppressor activity of CD4+CD25+Tregs, they were cocultured with CD4+CD25- effector T cells, and for T cell stimulation,anti-CD3/CD28 microbeads were applied alone or with Staphylococcus Enterotoxin B(SEB). The proliferation of the cells was measured by EZ4U colorimetric cell proliferationassay. Signifi cantly increased number of CD4+CD25brightFOXP3+ Tregs and also of CLA+CD4+CD25brightFOXP3+ Tregs were found in the peripheral blood of AD patients compared tohealthy individuals. The degree of suppressor activity of CD4+CD25+ regulatory T cells weredecreased in both anti-CD3/CD28 and SEB stimulated subgroups compared to control. Ourdata suggest coincidently with the literature that the pathogenesis of AD cannot be explainedwith the absence of CD4+CD25+FOXP3+ Treg cells, moreover the number of these cells isincreased in the peripheral blood. At the same time the function of Tregs may be impaired.

Megjegyzések:At the site of infl ammation infi ltrating CD4+ helper T cells play a pivotal role in the pathogenesis of atopic dermatitis (AD). IL-16 has been previously characterized as an immunomodulatory cytokine. Besides its chemotactic properties, IL-16 is able to trigger IL-2 receptor alpha chain expression and to activate CD4+ T cells, in synergy with IL-2 or IL-15. In addition, IL-16 amplifi es the infl ammatory processes by stimulating cytokine production in monocytes and contributes to T cell activation. Our aim was to investigate the association between serum IL-16 level and sensitisation in patients with AD. Overall 85 patients with AD were studied. Serum level of IL-16 was measured by immunoenzymatic assay in patients with AD and serum IgE, and specifi c IgE levels were also assessed. Based on specifi c IgE levels and Prick-test results, we divided AD patients into subgroups, and examined serum IL-16 and total IgE levels. Thirty-six healthy, aged-matched volunteers and 20 psoriasis patients served as controls.Compared to healthy individuals and patients with psoriasis, we found signifi cantly higher levels of IL-16 in sera of patients with AD. A signifi cant correlation was apparent between serum levels of IL-16 and the serum levels of IgE (r= 0.408; P=0.004). IL-16 levels were signifi cantly elevated in those subgroups, where specifi c IgE were detected against 1-5 or more than 5 antigens. Although the levels of IL-16 were also elevated in the Prick-test positive group, compared to Prick-test negative subjects, the difference was not signifi cant. IL-16 correlates with the degree of sensitization, but the association is not as strong as in the case of serum IgE. We assume that the role of IL-16 is complex in the pathogenesis of AD, both in the initiation and the perpetuation of the disease.

Megjegyzések:ackground Conflicting data exist on the number of invariantNKT (iNKT) cells in atopic dermatitis (AD); furthermore, nodata have been published on their functional capacity. Methods The frequency and number of circulatingCD3+6B11+ iNKT cells and their CD4+ and CD4?subpopulations were evaluated in peripheral blood obtainedfrom 41 patients with AD by four-color flow cytometry.Likewise, functional properties of iNKT cells were measuredby five-color intracellular cytokine staining. Results The number and percentage of total iNKT cells andtheir CD4/CD8 subpopulations were significantly lowerthan the controls. Of further importance, the CD4?CD8?(double negative, DN) iNKT subgroup showed thestrongest positive correlation with total iNKT cells. Inaddition, the DN subgroup exhibited the most pronouncedfunctional alteration with significantly decreasedlevels of intracellular IFN? and significantly increasedlevels of intracellular IL-4 in AD patients compared withthe controls. Conclusion The significantly altered number and cytokineproduction of iNKT cells from AD patients suggests thatthese cells may play an important role in the pathogenesisof AD.
