Cím:Regulatory T-cell subsets with acquired functional impairment : important indicators of disease severity in atopic dermatitis / Krisztián Gáspár, Sándor Baráth, Georgina Nagy, Gábor Mócsai, Edit Gyimesi, Péter Szodoray, Beatrix Irinyi, Margit Zeher, Éva Remenyik, Andrea Szegedi

Megjegyzések:Our aim was to assess whether the presence of highly active effector T-cells in atopic dermatitis (AD) is associated with changes in the number and/or function of regulatory T-cells (Tregs). Flow cytometry was utilised to determine the percentage of CD4+CD25brightCD127-/lowFOXP3+ and skin-homing CLA+CD4+CD25brightFOXP3+ Tregs in healthy controls and AD patients. The correlation between disease severity and Treg percentages was estimated. Treg suppressor activity and cell proliferation were measured after T-cell stimulation. Significantly increased percentages of Tregs were found in AD patients compared to healthy individuals, and significant correlation between the frequency of Tregs and disease severity was also detected. The otherwise normal suppressor activity of Tregs decreased in the presence of Staphylococcus enterotoxin B (SEB). In conclusion, the continuous presence of SEB can trigger an acquired functional impairment of Tregs in AD patients and the correlation between the increased frequency of Tregs and disease severity supports their important role in AD pathogenesis.

Cím:Association between serum IL-16 level and the degree of sensitization in patients with atopic dermatitis / Gáspár K., Nagy G., Irinyi B., Tumpek J., Sipka S., Remenyik É., Szodoray P., Szegedi A.

Megjegyzések:At the site of infl ammation infi ltrating CD4+ helper T cells play a pivotal role in the pathogenesis of atopic dermatitis (AD). IL-16 has been previously characterized as an immunomodulatory cytokine. Besides its chemotactic properties, IL-16 is able to trigger IL-2 receptor alpha chain expression and to activate CD4+ T cells, in synergy with IL-2 or IL-15. In addition, IL-16 amplifi es the infl ammatory processes by stimulating cytokine production in monocytes and contributes to T cell activation. Our aim was to investigate the association between serum IL-16 level and sensitisation in patients with AD. Overall 85 patients with AD were studied. Serum level of IL-16 was measured by immunoenzymatic assay in patients with AD and serum IgE, and specifi c IgE levels were also assessed. Based on specifi c IgE levels and Prick-test results, we divided AD patients into subgroups, and examined serum IL-16 and total IgE levels. Thirty-six healthy, aged-matched volunteers and 20 psoriasis patients served as controls.Compared to healthy individuals and patients with psoriasis, we found signifi cantly higher levels of IL-16 in sera of patients with AD. A signifi cant correlation was apparent between serum levels of IL-16 and the serum levels of IgE (r= 0.408; P=0.004). IL-16 levels were signifi cantly elevated in those subgroups, where specifi c IgE were detected against 1-5 or more than 5 antigens. Although the levels of IL-16 were also elevated in the Prick-test positive group, compared to Prick-test negative subjects, the difference was not signifi cant. IL-16 correlates with the degree of sensitization, but the association is not as strong as in the case of serum IgE. We assume that the role of IL-16 is complex in the pathogenesis of AD, both in the initiation and the perpetuation of the disease.

Results: In the total patient group, significantlyhigher levels of IL-16 were found in the sera of patientswith AD, compared to healthy individuals and patientswith psoriasis. A significant correlation was detected betweenserum levels of IL-16 and total IgE, total IgE andeosinophil counts, but not between IL-16 and eosinophils.
