Cím:IL-6 production of C2C12 cells is enhanced in the presence of macrophages and pravastatin / Karolina Cseri, Péter Szentesi, László Csernoch
Dátum:2021
ISSN:0231-5882 1338-4325
Megjegyzések:Skeletal muscle secrets several bioactive molecules known as myokines. Interleukin-6 (IL-6) has been described as a myokine secreted in response to skeletal muscle injury as well as to macrophage invasion in inflammation. To our knowledge no connection between macrophages and skeletal muscle regarding IL-6 secretion has been described so far. Here we report that co-culturing of C2C12 cells with RAW macrophages enhances IL-6 secretion of the cells cultured together. However, this is not seen in cross-feeding experiments, where culture medium of RAW macrophage culture is used as the culture medium of C2C12 cells or vice versa. Pravastatin, known to induce myopathy, also stimulates IL-6 production in monocultured C2C12 cells and elevates IL-6 concentration in the culture medium of the co-cultures. These results indicate an intricate interaction between skeletal muscle and macrophages in inflammation related to IL-6 production.

Cím:Caffeine and the myoplasmic calcium removal mechanisms in cut frog skeletal muscle fibres / Csernoch, L., Kovacs, L., Nilius, B., Szucs, G.
Dátum:1990
ISSN:0231-5882
Megjegyzések:Antipyrylazo III myoplasmic calcium transients were recorded in cut skeletal muscle fibres of the frog (Rana esculenta), using the double vaseline-gap voltage-clamp system. Intracellular calcium removal mechanisms were analysed, using a slightly modified model taken from the literature. Parameter values reported here are generally consistent with those obtained by the original model. Caffeine (0.5 mmol.l-1) moderately enhanced the overall myoplasmic calcium removal. In particular, the rate constant of the non-saturable uptake increased by 51% on the average, but there was a considerable fiber-to-fiber variation. The kinetic features of the binding sites representing the saturable uptake did not change significantly while the concentration of the available sites decreased by 36%. It is concluded that the caffeine-induced changes of the calcium removal components can be explained by supposing an increased resting myoplasmatic Ca2+ concentration in the presence of the drug.

Cím:Age-dependent changes in ion channel mRNA expression in canine cardiac tissues / Gönczi Mónika, Birinyi Péter, Balázs Bernadett, Szentandrássy Norbert, Harmati Gábor, Könczei Zoltán, Csernoch László, Nánási Péter P.
Dátum:2012
Megjegyzések:The expression pattern of cardiac ion channels displays marked changes during ontogeny. This study was designed to follow the developmental changes in the expression of major ventricularand atrial ion channel proteins (including both pore forming and regulatory subunits) in canine cardiactissues at the mRNA level using competitive reverse transcription polymerase chain reaction. Therefore, the corresponding mRNA levels were compared in myocardial tissues excised from embryonic (25?60days of gestation) and adult (2?3 years old) canine hearts. Expression level of Kv4.3, Kv1.4, KChIP2,KvLQT1, and Cav3.2 mRNAs were higher in the adult than in the embryonic hearts, while expressionof Nav1.5 and minK mRNAs were higher in the embryonic than in the adult myocardium. No change inKir2.1, HERG, Kv1.5, and Cav1.2 mRNA was observed during ontogeny. Direction of the developmentalchange in the mRNA level, determined for any specific channel protein, was identical in the atrial andventricular samples. The age-dependent increase observed in the expression of Kv4.3, Kv1.4, KChIP2, andKvLQT1 is congruent with the greater repolarization reserve of the adult myocardium, associated withhigher densities of Ito and IKs. The results indicate that age-dependent changes in the expression patternof many ion channels are similar in canine and healthy human myocardium, therefore, canine cardiacmuscle can be considered as a good model of studying developmental changes in the human heart.

Cím:Nicotine interferes with purinergic signaling in smooth muscle cells isolated from urinary bladders of patients with lower urinary tract symptoms / Ágnes Jenes, Gyula P. Szigeti, Ferenc Ruzsnavszky, Attila Varga, Laszlo Lorincz, Laszlo Csernoch
Dátum:2013
ISSN:1338-4325
Megjegyzések:In patients with outlet obstruction, the contraction of the base is reduced compared to that of healthy individuals, while the contraction of the dome is not affected. Here, we investigated the cellular mechanisms that might be responsible for cholinergic effects blocking non-adrenergic non-cholinergic contractions in the base of the urinary bladder. Smooth muscle cells either from the base or from the dome of human urinary bladders were cultured to determine the contribution of cholinergic and purinergic mechanisms to their Ca2+ homeostasis. While ATP evoked Ca2+ transients in all the cells, nicotine and carbachol induced Ca2+ transients only in 56% and 44% of the cells, respectively. When ATP was administered together with nicotine or carbachol, the amplitudes of the Ca2+ transients recorded from cells prepared from the base of bladders were significantly smaller (42 ± 6% with nicotine and 56 ± 9% with carbachol) than those evoked by ATP alone. This inhibition was much less apparent in the dome of bladders. The inhibition between the cholinergic and purinergic signaling pathways reported in this work may decrease the strength of the contraction of the base of the urinary bladder in patients with outlet obstruction during voiding.

ISSN:1338-4325
Megjegyzések:Very recently, the diamide insecticide chlorantraniliprole was shown to induce Ca2+-release from sarcoplasmic reticulum (SR) vesicles isolated from mammalian skeletal muscle through the activation of the SR Ca2+ channel ryanodine receptor. As this result raises severe concerns about the safety of this chemical, we aimed to learn more about its action. To this end, single-channel analysis was performed, which showed that chlorantraniliprole induced high-activity bursts of channel opening that accounts for the Ca2+-releasing action described before.

Cím:Laser induced calcium oscillations in fluorescent calcium imaging / János Vincze, Nikolett Geyer, Gyula Diszházi, László Csernoch, Tamás Bíró, István Jóna, Beatrix Dienes, János Almássy
Dátum:2018
Megjegyzések:Phototoxicity is the most common problem investigators may encounter when performing live cell imaging. It develops due to excess laser exposure of cells loaded with fluorophores and can lead to often overlooked but significant artifacts, such as massive increase of intracellular Ca2+ concentration, which would make data interpretation problematic. Because information about laserand dye-related changes in cytoplasmic calcium concentration is very limited, we aimed to describe this phenomenon to help investigators using laser scanning confocal microscopy in a non-invasive way. Therefore, in the present study we evaluated fluorescent fluctuations, which evolved in Fluo3/4/8 loaded mouse pancreatic acinar cells during very low intensity laser excitation. We demonstrate that after standard loading procedure (2 ?M Fluo-3/4/8-AM, 30 min @ room temperature), applying 488 nm laser at as low as ca. 10 ?W incident laser power (0.18 ?W/?m2) at 1 Hz caused repetitive, 2-3 fold elevations of the resting intracellular fluorescence. The first latency and the pattern of the fluorescence fluctuations were laser power dependent and were related to Ca2+-release from intracellular stores, as they were abolished by BAPTA-AM treatment in Ca2+- free medium, but were not diminished by the ROS scavenger DMPO. Worryingly enough, the qualitative and quantitative features of the Ca2+-waves were practically indistinguishable from the responses evoked by secretagogue stimulation. Since using similar imaging conditions, a number of other cell types were reported to display spontaneous Ca2+ oscillations, we propose strategies to distinguish the real signals from artefacts.
