Megjegyzések:OBJECTIVE: To investigate the release of arachidonic acid (AA) in unfractionated peripheral blood mononuclear cells (PBMC), separated monocytes and T lymphocytes of patients with systemic lupus erythematosus (SLE). METHODS: AA release was measured in cells from 56 patients with SLE and from 48 controls. Of the 56 patients with SLE, 38 were receiving glucocorticosteroids and 18 were not. [3H]AA was incorporated into the membranes of PBMC and purified subsets of monocytes and T lymphocytes. The release of [3H]AA was measured both in nonstimulated cells cultured for 24 h and in cell cultures stimulated by phorbol ester (PMA) and Ca2+ ionophore for 4 h. RESULTS: In the PBMC of SLE patients not taking glucocorticosteroids, the release of AA was decreased in both stimulated and nonstimulated cells. There was a decrease of AA production in monocytes but not in T lymphocytes. This phenomenon could be observed in the active and inactive phases of the disease. CONCLUSION: A defect in AA production may exist in the peripheral monocytes of patients with SLE, resulting in decreased release of AA in patients not receiving glucocorticosteroid therapy.

Megjegyzések:The objective of this study was to investigate the effects of heat shock (HS) treatment and geldanamycin (GA) on the release of arachidonic acid (AA) from human peripheral blood mononuclear cells (PBMC), monocytes and lymphocytes. Mononuclear cells prepared from blood of healthy subjects were preincubated with (3)H-AA. The release of (3)H-AA incorporated into the membrane was studied after pretreatment of cells by HS (43 degrees C, 1 h) and GA. The activation of AA producing enzymes was achieved by the addition of phorbol 12-myristate 13-acetate (PMA) or by the combination of PMA+calcium ionophore A-23187. Treatment of cells by HS inhibited the release of AA. Furthermore, the release of AA by PBMC was dose dependently inhibited by GA. The combination of treatments by HS and GA augmented the inhibition of AA release. The HS response involves a diminished release of AA from PBMC. The inhibitory effect of GA on the AA release is a new element in the antiinflammatory pharmacological ability of this drug.

Megjegyzések:The changes in the number of CD8(+) T lymphocytes were studied before (0 day) and then 30 days after the autologous hematopoietic stem cell transplantations (AHSCT) in 14 therapy refractory patients with autoimmune diseases. The years of survival and the clinical states were also evaluated. The number of CD8(+) T cells was determined by an hematologic automat and by flow cytometry. Longer than 5-year survival times were found in 6 cases, whereas there was no progression (improvement) in 2 cases, and 4 patients were lost. The increase in the number of CD8(+) cytotoxic T cells was gradual in the first 2 months and reached the significantly highest values among all subtypes of lymphocytes. It was of a special interest that in all the 4 patients who died, the numbers of CD8(+) T cells were less than 150/?l on the 30th day after AHSCT, whereas all the 10 patients with a higher cell number survived. These results suggest that the early monitoring of the number (not only the ratio) of regenerating CD8(+) T cells in the peripheral blood can be a useful and quantitative laboratory measurement after AHSCT, and it has a significant relation also to the survival times of transplanted patients.
