Megjegyzések:Radioimmunoassay (RIA) methods for ethynylestradiol in body fluids usually take more than 24 hours. This paper describes a rapid RIA procedure developed by the authors for determination of ethynylestradiol. Ethynylestradiol antiserum was used in a dilution of 1:8000 (corresponding to a final dilution of 1:40,000) and its cross reactivity determined. With 17 beta-estradiol, the antiserum exhibited 5.54% cross-reaction. Separation of the free and bound hormones was done using 0.1% Dextran T 70-coated 0.625% Norit A charcoal. The scintillation and extraction procedures are described. Sensitivity of the rapid RIA method was found to be 28.2 pmol/1. Its precision (n=50) was established: the intra-assay coefficient of variation was 4.5%, and the inter-assay coefficient of variation 8%. Blank experiments performed showed that water, male serum, and the serum of nonpregnant women who were not taking contraceptive pills gave values of 60.7 +or- 16.9 pmol/l, 64.1 +or- 23.6 pmol/l, and 74.2 +or- 10.1 pmol/l, respectively. These values are relatively low and insignificant compared to the serum level of women taking ethynylestradiol-containing pills. Serum ethynylestradiol 2 level was 270 +or- 85 pmol/l 6-12 hours following oral administration of 0.05 mg hormone.
