Megjegyzések:We examined whether the inhibition ofnitric oxide (NO) synthesis by NG-nitro-L-arginine (LNNA) abolished pacing-induced preconditioning, and ifprolonged exposure to cholesterol-enriched diet led to the loss of preconditioning due to decreased cardiac NOformation. Therefore, Wistar rats fed 2% cholesterol-enriched diet or standard diet for 24 weeks were treatedwith a single dose of 1 mg/kg LNNA or its solvent at the end of the week 24, respectively. Isolated hearts fromall groups were subjected to either preconditioning induced by three consecutive periods of pacing at 600 beats/min for 5 min, with 5-min interpacing periods, or time-matched non-preconditioning perfusion, followed by a10-min coronary occlusion, respectively. In the control group, coronary occlusion after a non-preconditioningprotocol decreased aortic flow (AF) from 45.4?2.4 to 15.6?1.5 ml/min, and resulted in a lactate dehydrogenase(LDH) release of 219?55 mU/min/g, however, preconditioning attenuated the consequences of coronary occlusion[AF: 27.3?1.7 ml/min (P<0.05); LDH: 44?14 mU/min/g (P<0.05)]. Preconditioning did not confer protectionin the LNNA-treated (AF: 17.4?1.5 ml/min; LDH: 151?21 mU/min/g), and/or in the high-cholesterol-fed groups(AF: 15.7?1.2 ml/min; LDH: 168?22 mU/min/g). Preconditioning was preserved however, when hearts weretreated with LNNA after the preconditioning protocol [AF: 29.6?2.2 ml/min (P<0.05); LDH: 48?17 mU/min/g (P<0.05)]. Both LNNA treatment and cholesterol-enriched diet markedly decreased cardiac NO content assayedby electron spin resonance spectroscopy. We conclude that NO may be involved in the triggering mechanism ofpacing-induced preconditioning, the protective effect of which is blocked by sustained exposure to dietarycholesterol, possibly by influencing cardiac metabolism of NO.

Megjegyzések:AIM: The sphincter of Oddi (SO) plays an important role indelivery of bile into the duodenum. To establish whethervasoactive intestinal polypeptide (VIP) and nitric oxide (NO)were involved in phasic contractile activity of the rabbitSO stimulated by cholecystokinin-octapeptide (CCK-8).METHODS: Isolated SO muscle rings were cleaned of fatand mounted horizontally on two small L-shaped hooksone of which was connected to a force transducer for themeasurement of isometric tension. The experiments werecarried out in a thermostatically controlled (37?0.2 )organ bath (5 mL) containing Krebs solution. The organfluid was gassed with 95% O2 and 50 mL/L CO2 to keepthe pH at 7.40?0.05. Contractile responses to CCK-8(1 ?mol/L) were evaluated in the presence and absenceof NG-nitro-L-arginine (LNNA), an inhibitor of NO synthase(100 ?mol/L), and (p-chloro-D-Phe6-Leu17)-VIP (VIPa,30 ?mol/L), a VIP receptor antagonist.RESULTS: CCK-8 stimulated the phasic activity of the SO.NO synthase inhibition increased the frequency and amplitudeof contractions with a slight increase in developed tension.Pre-incubation with VIPa also attenuated this CCK-8 effect.The combined application of LNNA and VIPa abolishedthe phasic activity of the muscle rings with a marked increasein tension in response to CCK-8.CONCLUSION: VIP and NO together contribute to anincrease in phasic activity of SO.

We studied whether non-adrenergic, non-cholinergic (NANC) relaxation of the rabbit sphincter of Oddi was influenced by tolerance to nitroglycerin (NG)in vitro. Sphincter of Oddi (SO) muscle rings precontracted with EC50concentrations of cholecystokinin octapeptide (CCK8) were exposed to cumulative increases in NG concentrations and tested for relaxation by measurement of isometric tension. A separate group of six rings was subjected to a preceding exposure to 275 mnitroglycerin over 60 min to inducein vitrotolerance to nitroglycerin. The rings (both tolerant and non-tolerant) were subjected to electrical field stimulation (FS: 50 V, 0.1 ms, 20 Hz, 3 and 10 stimuli). The rings were then preincubated with NANC solution: phentolamine, oxprenolol and atropine (all 1 m) for 20 min and FS was applied again. FS was repeated after additional incubation withNG-nitro-l-arginine methyl ester (l-NAME), an inhibitor of NO synthase (30 m) and after a successive incubation with 3 mml-arginine (20 min). Maximum contractions produced by CCK8 in `tolerant' and 'non-tolerant' sphincters were 29.9±5.8 and 28.3±5.2 mN, respectively. The sensitivity to CCK8 also was not different between the two groups with EC50(-log M) values of 8.5±0.2 and 8.3±0.1, respectively. FS evoked twitchlike contraction followed by relaxation in the ampullary SO in both `tolerant' and `non-tolerant' preparations. Incubation in NANC solution resulted in monophasic relaxations in response to FS in non-tolerant sphincters but not in tolerant ones.l-NAME (30 m) reversed NANC relaxation in non-tolerant muscle rings whereas it failed to modify NANC contractions in the tolerant preparations.l-arginine (3 mm) reversed the inhibitory effect ofl-NAME on NANC relaxation in the `non-tolerant' rings and it was without effect on FS-induced contractions in the `tolerant' SO. As measured by radioimmunoassay, tolerance to NG was without any significant effect on tissue content of both cyclic adenosine 3:5 monophosphate (cAMP) and cyclic guanosine 3:5 monophosphate (cGMP). FS significantly increased tissue cAMP and cGMP content in 'non-tolerant' preparations. FS failed to increase the level of either cyclic nucleotide in `tolerant' tissue. We conclude that NANC relaxation of the ampullary part of the rabbit SO is significantly impaired in the state of tolerance to NG`in vitro'.

Megjegyzések:Multiple brief periods of rapid ventricular pacing confer bothshort- and long-term protection on the ischaemic heart. The duration of the short-term protection does notexceed 2 h, whereas the long-term protective effect appears several hours after the inducing insults, with maximalprotection 24?48 h later. Up to now, delayed cardiac protection by preceding ischaemic insults against harmfulconsequences of stress has been produced in the normal, healthy animal only. The purpose of this study was,therefore, to test whether delayed cardiac protection can be induced in experimental atherosclerosis in rabbitsproduced by feeding cholesterol-rich diet over 2 months. Repeated brief periods of rapid ventricular pacing wereused to induce delayed protection of the heart. Moderation of post-pacing right intracavitary ST segment elevationand that of the left ventricular end-diastolic pressure (both produced by ventricular overpacing: 500 beats/minfor 15 min) were found in normal animals as well as in those fed cholesterol-enriched diet. The short-livedprotection induced by a single 'preconditioning' pacing was reproducible only in normal animals. As measuredby means of radioimmunoassay, the protective effect of either short- or long-term protection appeared inparallel with an attenuation of ischaemia-induced increase in cardiac cyclic AMP content, in both normal andatherosclerotic rabbits. An increase in cardiac cyclic GMP content was characteristic of the short- but not longtermprotection. These results suggest that the delayed cardiac protection by preceding multiple brief rapidpacings operates even in experimental atherosclerosis, but the short-term protection induced by a singlepreconditioning stimulus is lost.

Megjegyzések:In this article we studied whether the nitrergic relaxation of the rabbit sphincter wassensitive to glibenclamide. Field stimulation relaxed the sphincter of Oddi rings afterincubation with atropine 1 mM. and guanethidine 4 mM. with threefold and fourfoldincreases in tissue guanosine 39:59-cyclic monophosphate and adenosine 39:59-cyclicmonophosphate contents, respectively. These changes were blocked by 30 mM NG-nitro-L-arginine methyl ester. Glibenclamide 0.1]10 mM. attenuated the field stimulation-inducedrelaxation and completely abolished the relaxation produced by 0.1 mM cromakalim. Weconclude that nitrergic relaxation of the rabbit sphincter of Oddi comprises a mechanismsensitive to glibenclamide.

Megjegyzések:Field stimulation relaxed the sphincter of Oddi muscle rings of the rabbit after incubation with phentolamine, oxprenolol and atropine (all 1 microM). The relaxation was blocked by NG-nitro-L-arginine methyl ester (30 microM) and was reversed by 3 mM L-arginine but not D-arginine. Sphincter of Oddi preparations from hypercholesterolaemic rabbits exhibited contractions under the same conditions. We conclude that nitrergic relaxation is impaired in the sphincter of Oddi from hypercholesterolaemic rabbits.

Megjegyzések:A preceding right ventricular overdrive pacing (VOP) of 500 b.p.m. for 5 min, markedly reduced the severity of global myocardial ischaemia produced by a subsequent 5-min VOP in conscious rabbits. This VOP-induced preconditioning developed in parallel with an increase in cardiac cyclic guanosine 3':5'-monophosphate (cyclic GMP) content. VOP-induced preconditioning was abolished when the animals had been made tolerant to the vasodilator effect of nitroglycerin (NG). In the heart of the NG-tolerant rabbits, neither VOP nor preconditioning increased cyclic GMP content. This suggests that changes by NG tolerance of cyclic GMP metabolism may account for the loss of VOP-induced preconditioning.

Megjegyzések:A brief rapid pacing has been shown to protect rabbit heart against global myocardial ischaemia induced by subsequent longer pacing. We studied whether pacing-induced preconditioning was reproducible in experimental hypercholesterolaemia. In conscious rabbits with an implanted right ventricular electrode and left ventricular polyethylene catheters, pacing of 500 bpm over 20 min induced an intracavitary ST-segment elevation of 3.2 +/- 0.41 mV, shortened ventricular effective refractory period and increased left ventricular end-diastolic pressure from prepacing 105 +/- 3.9 ms and 4.0 +/- 0.93 mmHg to post-pacing 62 +/- 6.4 ms and 27.9 +/- 7.2 mmHg, respectively. A 10-min preconditioning pacing followed by a 5-min interval markedly attenuated these test pacing-induced ischaemic changes. Rabbits were fed a cholesterol-enriched diet over 4, 8 and 12 weeks, responded to a 5- or 10-min pacing with ischaemic changes of the same degree as did controls to a 10- or 20-min pacing, respectively. A 4-week diet elevated total serum cholesterol from 1.7 +/- 0.4 to 24.1 +/- 2.9 mmol/l without apparent atherosclerotic lesions in the thoracic aorta assessed by Oil-Red O staining and planimetry, but it abolished protection induced by a 5-min preconditioning pacing. A 12-week diet increased serum cholesterol and lesion surface area to 26.9 +/- 3.2 mmol/l and 89.6 +/- 6.4%, respectively, and continued to block preconditioning. When these animals were refed normal chow over additional 6 weeks, serum cholesterol level dropped to 2.6 +/- 0.80 mmol/l with no change in atherosclerotic lesions, the preconditioning effect, however, recovered. We conclude that hypercholesterolaemia blocks preconditioning irrespective of the development of atherosclerosis.

Megjegyzések:AbstractField stimulation relaxed the rabbit sphincter of Oddi muscle rings after incubation with atropine (1 microM) and guanethidine (4 microM) with a threefold increase in tissue cyclic cGMP content, a response previously shown to be essentially nitrergic. Preparations from hypercholesterolaemic rabbits (1.5% dietary cholesterol load over 8 weeks increasing serum total cholesterol from pre-diet 1.4+/-0.3 to 22.6+/-3.8 mmol/l) exhibited contractions with no change in cyclic GMP content under the same conditions. The nitrergic relaxation was recaptured with a twofold increase in tissue cyclic GMP content in preparations from hypercholesterolaemic rabbits undergone a treatment with 30 microM/kg farnesol i.v. twice a day over the last 3 days of the dietary period. We conclude that farnesol treatment restores nitrergic relaxation of the sphincter of Oddi in hypercholesterolaemia.
