Megjegyzések:The number of circulating platelets dropped abruptly in the early phase of severe anaphylactic shock (AS) of the rat and could be inhibited by Persantin but not by heparin pretreatment. The results strongly suggest that an aggregating agent, perhaps "platelet aggregating factor", formed or released during anaphylaxis is responsible for the decrease of platelet number. The organ distribution of 51Cr labelled platelets showed that in AS the aggregated platelets were removed from the circulation mostly by the spleen and part of them were trapped in the lung and the small intestine. The remaining platelets retained their functional integrity. The loss of circulating platelets is manifested by the prolongation of rat tail bleeding time and is one of the factors participating in the haemostatic disturbances during anaphylaxis.

Megjegyzések:Attempts were made to detect soluble immune complexes (IC) in sera of rats submitted to anaphylactic shock 12 days following BPV plus antigen treatment, and their in vitro formation following the addition of specific antigens to sera or heparinized platelet rich plasma of sensitized rats. We failed to detect ICs or IC formation by studying Hageman factor activation and platelet aggregation or by using a very sensitive radio-assay of ICs. It is established that the anaphylactic activation of Hageman factor as well as the aggregation of platelets are not mediated by immune complexes, therefore, other explanations must be found.

Megjegyzések:Four fractions, fraction A,B, C, and D, were isolated from the aqueous extract of wheat flour on a Sephadex G-75 column. It was found that the repeated gel-filtrated fraction A reacts as an antigen with the sera of coeliac patients. On the basis of its molecular weight and amino acid compositions, it is gliadin. Each of the 28 coeliac patients had heat-stable precipitating antibody and higher than normal IgE levels were found in the sera of only three individuals. Specific antigliadin IgE in low quantity was found in only one case. The sera reacting with antigens in high titres were absorbed by anti-IgG and anti-IgA whereupon the antibody level considerably decreased. The main circulating antibody of the patients was IgG.
