Glucocorticoids are powerful anti-inflammatory compounds inhibiting inflammatory cell recruitment and production of pro-inflammatory cytokines. We have recently found that dendritic cells, the key-players of T cell priming and polarization respond to allogeneic apoptotic neutrophils with proinflammatory cytokine release and Th1 cell activation. Here we show that monocyte-derived human dendritic cells develop their capacity to engulf apoptotic cells by up-regulating a set of apopto-phagocytic genes. This gene expression pattern was reprogrammed when differentiation took place in the presence of the synthetic glucocorticoid dexamethasone (Dex) which increased the expression of phagocytosis receptors MERTK and CD14, the bridging molecule C1QA, DNASE2 and the adenosine A3 receptor (ADORA3). The increased phagocytosis was attenuated by the addition of ADORA3 antagonist and could not be observed when bone marrow derived dendritic cells of ADORA3 knockout mice were treated with Dex. The glucocorticoid treated human dendritic cells loaded with allogeneic apoptotic neutrophils secreted, in response to LPS and IFN, the inflammatory cytokine TNFα. Furthermore, the Dex treated dendritic cells could activate autologous T lymphocytes toward Th1 effector cells and this was enhanced by their exposure to allogeneic apoptotic neutrophils.

The daily clearance of billions of physiologically dying cells in the human body is performed safely mainly by the mononuclear phagocyte system Bridging molecules and receptors ? which recognize the dying cells ? show high redundancy and are linked to several intracellular signaling pathways. Our previous ♭Taq-Man Low Density Array' measurements predicted important role of many genes in phagocytosis, because their expression level heavily elevated during the early stage of the phagocytosis process. The four genes (ADORA2A, FPRL1, ICAM3, THBS1)with the most enhanced expression in human macrophages wereknocked-down by RNA interference which was controlled byWestern-blot and surface immunostaining at translational level. Significant decrease in phagocytic capacity was observed only after silencing ICAM3. Our goal was to investigate the role and interacting partners of ICAM3 transmembrane protein from both sides. Human monocytes were isolated from ♭buffy coats' of healthy blood by magnetic separation using CD14 human microbeads. To examine the phagocytic capacity of 5 day differentiated macrophages, apoptotic neutrophil granulocytes were isolated from human blood by Histopaque density-gradient centrifugation. The phagocytosis assay was performed using fluorescent labeled cells and the incorporated cell-rate was measured by flow cytometry, immediately after pre-incubation of macrophages or apoptotic cells with blocking antibodies. Significant reduction of phagocytosis was noticed after blocking ICAM3 from both sides while that of DC-SIGN, which is a cited receptor for ICAM3, did not influence the engulfment of apoptotic cells. ICAM-3 participates in the recognition of apoptotic neutrophils by macrophages from both sides not only as an adhesion molecule but also as an initiator of signaling pathways mediated by intracellular thyrosin kinases. To examine its interacting partners in this process further investigations have to be performed in the future.

Apoptotic cells express eat-me signals which are recognized by several receptors mainly on professional phagocytes that can define their safe and effective clearance in order to maintain tissue homeostasis in the entire body. TLDA measurements predicted important role of some apopto-phagocytic genes in phagocytosis of apoptotic neutrophils by macrophages, because their expression level heavily elevated during the early stage of phagocytosis. After they were silenced by RNA interference, significant decrease in phagocytosis was observed only after silencing ICAM3. Our goal was to investigate the role and interacting partners of ICAM3 from both sides. Monocytes were isolated from buffy coats of healthy blood by CD14 specific magnetic separation. To examine phagocytic capacity of differentiated macrophages, apoptotic neutrophils were isolated from human blood by density-gradient centrifugation. Phagocytosis assay was performed using fluorescent labeled cells and the incorporated cell-rate was measured by flow cytometry, immediately after preincubation of macrophages or apoptotic cells with blocking antibodies. Localization of the investigated receptors was visualized by indirect immunostaining. Significant reduction of phagocytosis was noticed after blocking of ICAM3 from both sides. In macrophages but not in neutrophils silencing and blocking components of LFA-1, which can strongly bind ICAM3, resulted in a decreased phagocytosis of apoptotic cells. Engulfing caps formed in macrophages during phagocytosis are characterized by accumulation of ICAM3 and the subunits of LFA-1 which show colocalization on the surface of phagocytes showing that ICAM3 and LFA-1 act as recognition receptors in the phagocytosis portals of macrophages for engulfment of apoptotic neutrophils.

Apoptotic cells express eat-me signals which are recognized by several receptors mainly on professional phagocytes of the mononuclear phagocyte system. This "engulfment synapse" can define a safe and effective clearance of apoptotic cells in order to maintain tissue homeostasis in the entire body. We show that the expression of four genes related to apoptotic cell clearance is strongly up-regulated in human macrophages 30 min after administration of apoptotic neutrophils. Out of these the significant role of the up-regulated intercellular adhesion molecule 3 (ICAM3) in phagocytosis of apoptotic neutrophils could be demonstrated in macrophages by gene silencing as well as treatment with blocking antibodies. Blocking ICAM3 on the surface of apoptotic neutrophils also resulted in their decreased uptake which confirmed its role as an eat-me signal expressed by apoptotic cells. In macrophages but not in neutrophils silencing and blocking integrin alphaL and beta2 components of lymphocyte function-associated antigen 1 (LFA-1), which can strongly bind ICAM3, resulted in a decreased phagocytosis of apoptotic cells indicating its possible role to recognize ICAM3 on the surface of apoptotic neutrophils. Finally, we report that engulfing portals formed in macrophages during phagocytosis are characterized by accumulation of ICAM3, integrin alphaL and beta2 which show co-localization on the surface of phagocytes. Furthermore, their simultaneous knock-down in macrophages resulted in a marked deficiency in phagocytosis and a slight decrease in the anti-inflammatory effect of apoptotic neutrophils. We propose that ICAM3 and LFA-1 act as recognition receptors in the phagocytosis portals of macrophages for engulfment of apoptotic neutrophils.

MCF-7 cells undergo autophagic death upon tamoxifen treatment. Plated on non-adhesive substratum these cells died by anoikis while inducing autophagy as revealed by monodansylcadaverine staining, elevated light-chain-3 expression and electron microscopy. Both de novo and anoikis-derived autophagic dying cells were engulfed by human macrophages and MCF-7 cells. Inhibition of autophagy by 3-methyladenine abolished engulfment of cells dying through de novo autophagy, but not those dying through anoikis. Blocking exposure of phosphatidylserine (PS) on both dying cell types inhibited phagocytosis by MCF-7 but not by macrophages. Gene expression profiling showed that though both types of phagocytes expressed full repertoire of the PS recognition and signaling pathway, macrophages could evolve during engulfment of de novo autophagic cells the potential of calreticulin-mediated processes as well. Our data suggest that cells dying through autophagy and those committing anoikis with autophagy may engage in overlapping but distinct sets of clearance mechanisms in professional and non-professional phagocytes

The daily clearance of physiologically dying cells is performed safely mainly by cells in the mononuclear phagocyte system. They can recognize and engulf dying cells utilizing several cooperative mechanisms. In our study we show that the expression of a broad range of apopto-phagocytic genes is strongly up-regulated during differentiation of human monocytes to macrophages with different donor variability. The glucocorticoid dexamethasone has a profound effect on this process by selectively up-regulating six genes and down-regulating several others. The key role of the up-regulated mer tyrosine kinase (Mertk) in dexamethasone induced enhancement of phagocytosis could be demonstrated in human monocyte derived macrophages by gene silencing as well as blocking antibodies, and also in a monocyte-macrophage like cell line. However, the additional role of other glucocorticoid induced elements must be also considered since the presence of autologous serum during phagocytosis could almost completely compensate for the blocked function of Mertk.
