Megjegyzések:Rationale and Objectives. The malignant hyperthermia (MH) syndromeinvolves characteristic changes in metabolism that have a significanteffect on the postmortem properties of muscle tissue. We investigatedthe H-1 nuclear magnetic resonance (MR) relaxation parameters of muscletissue from normal (MH-) and malignant hyperthermia-susceptible (MH+)swine taken before their death to identify whether postmortem metabolicdifferences would be accompanied by changes in relaxation parameters.Methods. Muscle samples from 63 MH-susceptible swine were characterizedby proton MR relaxation times (T1 and T2). The water content, pH,conducting, and the ''subjective meat'' quality were assessed in orderto differentiate the meat into one of three groups: normal,pale-soft-exudative, and dark-firm-dry.Results. The T2 relaxation of MH- and MH+ muscles was significantlydifferent, but no significant changes were detected in the watercontent and corresponding T1 relaxation times.Conclusion. These data suggest that MH-susceptible swine can bedistinguished from normal swine using nuclear MR relaxometry. Theresults of relaxation measurements and the multiexponential analysis ofthe T2 curves suggest a shift between the different water compartmentsduring MH that might predict the increased water loss of muscle tissueafter death.

Megjegyzések:The Gd3+ complexes of three new phosphorus containing tetraazamacrocycles (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetrakis(methylene ethylphosphonic acid), H4DOTEP;1,4,7,10-tetraazacyclododecane-1,4,7,10-tetrakis (methylene phosphonicacid monoethylester), H4DOTPME; and the corresponding n-butyl ester,H4DOTPMB) were prepared and examined for possible use as MRI contrastagents. Although thermodynamically and kinetically less stable thanGd(DOTA)- in saline and HSA solution, the stability of these newmacrocyclic complexes appears to be sufficiently high for In vivoapplications. NMRD relaxivity profiles of the three complexes indicatethat the number of inner sphere water molecules for these chelates isless-than-or-equal-to 1 and that the more hydrophobic chelate,Gd(DOTPMB), binds to human serum albumin (HSA). Biodistribution studiesof the radioactive Sm-153 or Gd-159 chelates in rats, gamma imaging ofthe Sm-153 chelates in rats, and proton MRI studies of thenonradioactive. Gd3+ chelates in rabbits all indicate that the DOTPMBcomplexes accumulate preferentially in the liver, spleen, and smallintestines while the more hydrophilic DOTEP and DOTPME complexes appearto display renal clearances similar to other low molecular weightcontrast agents.

Megjegyzések:After months of restrictive containment efforts to fight the severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) epidemic, European countries are planning to reopen. To support the process, we conducted a cross-sectional survey among the Hungarian population to estimate the prevalence of infectious cases and prior SARS-CoV-2 exposure. A representative sample (n = 17,787) for the Hungarian population of 14 years or older living in private households (n = 8,283,810) was selected. The study was performed within 16 days after 50 days of restrictions, when the number of confirmed cases was stable low. Naso- and oropharyngeal smears and blood samples were collected for PCR and antibody testing. The testing was accompanied by a questionnaire about symptoms, comorbidities, and contacts. Design-based prevalence estimates were calculated. In total, 10,474 individuals (67.7% taken into account a sample frame error of 2315) of the selected sample participated in the survey. Of the tested individuals, 3 had positive PCR and 69 had positive serological test. Population estimate of the number of SARS-CoV-2 infection and seropositivity were 2421 and 56,439, respectively, thus active infection rate (2.9/10,000) and the prevalence of prior SARS-CoV-2 exposure (68/10,000) was low. Self-reported loss of smell or taste and body aches were significantly more frequent among those with SARS-CoV-2. In this representative, cross-sectional survey of the Hungarian population with a high participation rate, the overall active infection rate was low in sync with the prevalence of prior SARS-CoV-2 exposure. We demonstrated a potential success of containment efforts, supporting an exit strategy.

Megjegyzések:Intracellular K+/Na+ ratios of erythrocytes of various mammalianspecies are known to differ markedly. We have measured ATP, K+, Na+,Mg2+, H2O contents of erythrocytes of twelve mammalian and three avianspecies. Our results indicate that the intracellular ATP concentrationin erythrocytes of different species is in close positive correlationwith the K+/Na+ ratios (linear correlation coefficient, r = 0.852).Furthermore, ATP levels in erythrocytes of individual sheep withdifferent potassium concentrations correspond with their K+/Na+ ratios(r = 0.747). Intracellular magnesium concentrations also correlate withATP concentrations in erythrocytes of different species (r = 0.629) andin different sheep (r = 0.549).

These differences are sustaineddespite plasma potassium and sodium concentrations being almostidentical, i.e. high [Na+] and low [K+], in all mammalian species.Conventional cell theory couples intracellular K+ and Ne concentrationswith Na+/K+-dependent ATPase activity. Ling's Association-Induction(AI) hypothesis couples intracellular K+ and Na+ concentrations withthe concentration of ATP, by way of an inductive adsorption of bothmonovalent cations on the available terminal and lateral carboxylgroups of proteins. Based on the working hypothesis, differentrelationships are expected between the erythrocyte K+ and Na+concentrations and ATP levels.Our results indicate that the intracellular ATP level in erythrocytesof different species corresponds with intracellular K+ and Ne levels,K+/Ne ratios and total K+ + Na+ levels. Similar relationships wererevealed between the erythrocyte ATP and monovalent cation contents ofdifferent individual sheep.
