Megjegyzések:For the purpose of targeted drug delivery,composite biodegradable nanoparticles were prepared fromchitosan and the poly-?-glutamic acid via an ionotropicgelation process. These stable self-assembled nanoparticleswere characterized by dynamic light scattering, transmissionelectron microscopy, and atomic force microscopy,which demonstrated that the nanosystem consists ofspherical particles with a smooth surface both in aqueousenvironment and in dried state. Toxicity measurementsshowed that the composition is nontoxic when tested eitheron cell cultures or in animal feeding experiments. Toevaluate the potential of the nanosystem for intracellulardrug delivery, the nanoparticles were fluorescently labeledand folic acid was attached as a cancer cell-specifictargeting moiety. The ability of the particles to beinternalized was tested using confocal microscopic imagingon cultured A2780/AD ovarian cancer cells, which overexpressfolate receptors. The quantitative data obtained bydigital processing of the intensity of green color of eachpixel in the pictures inside the cell boundaries and totalintensity of fluorescence inside the cells showed that"targeted" particles internalized into the cells significantlyfaster and the total accumulation of these particles wassubstantially higher in the cancer cells when compared with"nontargeted" particles, which may facilitate effective andspecific cytoplasmic delivery of anticancer agents loadedinto such nanoparticles.
