Megjegyzések:White oaks (Quercus section, Quercus subgenus) are widely distributed in Europe. Quercus petraea (sessile oak), an economically important species is predicted to be affected by climate change. Q. pubescens (pubescent oak) and Q. virgiliana (Italian pubescent oak) are economically less important, drought tolerant species. Frequent hybridization of white oaks was observed and currently the introgression of Q. pubescens and Q. virgiliana in non-mediterranean regions of Europe has been reported. Our goal was to use tissue cultures established from individual trees of the above taxa and their putative hybrids, all present in the forest stand of Síkfőkút LTER Research Area (NE Hungary) as simple experimental model systems for studying drought/osmotic stress tolerance. Tissue cultures are more suitable models for such studies, than seedlings, because they are genetically identical to the parent plants. Polyethylene glycol (PEG6000) treatments were used for this purpose. The identification of taxa was based on leaf morphological traits and microsatellite analysis and showed that Q. petraea is genetically distinct to all other taxa examined. We established six callus lines of Quercus. As expected, in Q. petraea cultures PEG6000 induced severe loss of fresh weight and the ability to recover after removal of the osmoticum, which was not characteristic for Q. pubescens and Q. virgiliana. Putative hybrids exhibited an intermediate response to osmotic stress. Activity gels showed the increase of single-strand preferring (SSP) nuclease and no significant change of guaiacol-peroxidase activities in drought-sensitive genotypes/cultures and no significant increase of SSP nuclease activities accompanied with increases of guaiacol-peroxidase activities in drought-tolerant ones. This indicates that drought/osmotic stress tolerance is associated to increased capacity of scavenging reactive oxygen species and hence less susceptibility to DNA damage. Our results confirm that tissue cultures of oak are suitable model systems for studying drought/osmotic stress responses.

Megjegyzések:Turkey oak (Quercus cerris L.) is one of the most ecologically and economically significant deciduous tree species in the Central and Southeast European regions. The species has long been known to exhibit high levels of genetic and phenotypic variation. Recent climate response predictions for Turkey oak suggest a significant distribution extension in Europe under climate change. Since Turkey oak has relative drought-tolerant behaviour, it is regarded as a potential alternative for other forest tree species during forestry climate adaptation efforts, not only in its native regions but also in Western Europe. For this reason, the survey of existing genetic variability, genetic resources, and adaptability of this species has great significance. Next-generation sequencing approaches, such as ddRAD-seq (Double digest restriction-site associated DNA sequencing), allow the attainment of high-resolution genome-wide single nucleotide polymorphisms (SNPs). This study provides the first highly variable genome-wide SNP data for Turkey oak generated by ddRAD-seq. The dataset comprises 17 607 de novo and 26 059 reference mapped SNPs for 88 individuals from eight populations, two from Bulgaria, one from Kosovo, and five from Hungary. Reference mapping was carried out by using cork oak's (Quercus suber L.) reference genome. The obtained high-resolution genome-wide markers are suitable for investigating selection and local adaptation and inferring genetic diversity, differentiation, and population structure.

Megjegyzések:This paper analyses the drought-induced growth responses of two ecologically and economically important broadleaf tree species, namely sessile oak (Quercus petraea (Matt.) Liebl.) and Turkey oak (Quercus cerris L.) in south-western Hungary. Although both have been described as drought-tolerant species, our understanding of their growth responses to drought extremes is still limited. Forest stands of younger ages of the target species were sampled along a precipitation gradient under similar soil conditions. 136 tree-ring samples were used to build six species- and site-specific chronologies. We also applied several dendroecological metrics to assess the drought sensitivity of the species. Water deficit was estimated by various drought indices including the soil water-budget based water stress index. The results indicated a strong dependency of annual tree-ring width of both species on the water availability of summer months in the actual year of ring formation. However, we found markedly different reaction of the two species against drought conditions. Turkey oak responded more sensi- tively to droughts than sessile oak revealed by the significantly lower resistance and higher recovery potential of this species. The more plastic behaviour of Turkey oak could be also perceived by the higher sensitivity of its tree-ring chronologies and by the observed relationship between resistance and recovery that showed a closer fit to the line of full resilience in case of this species. Regarding the precipitation gradient, a linearly proportional increase of growth reduction with rising water stress was found for Turkey oak while the growth response of sessile oak decreased considerably with increasing aridity indicating lower growth plasticity of sessile oak to droughts there. Based on our findings it seems that Turkey oak copes better with droughts than sessile oak and may gain competitive advantages under the projected climate change in Hungary
