Megjegyzések:Sugar-modified nucleosides are prime synthetic targets in anticancer and antiviral drug development. Radical mediated thiol?ene coupling was applied for the first time on nucleoside enofuranoside derivatives to produce a broad range of thio-substituted D-ribo, -arabino, -xylo and L-lyxo configured pyrimidine nucleosides. In contrast to the analogous reactions of simple sugar exomethylenes, surprisingly, hydrothiolation of nucleoside alkenes under the standard conditions of various initiation methods showed low to moderate yields and very low stereoselectivity. Optimizing the reaction conditions, we have found that cooling the reaction mixture has a significant beneficial effect on both the conversion and the stereoselectivity, and UV-light initiated hydrothiolation of C2·-, C3·- and C4·-exomethylene derivatives of nucleosides at ?80 ?C proceeded in good to high yields, and, in most cases, in excellent diastereoselectivity. Beyond the temperature, the solvent, the protecting groups on nucleosides and, in some cases, the configuration of the thiols also affected the stereochemical outcome of the additions. The anomalous L-lyxo diastereoselectivity observed upon the addition of 1-thio-?-D-gluco- and galactopyranose derivatives onto C4·,5·-unsaturated uridines is attributed to steric mismatch between the D-ribo C4·-radical intermediates and the ?-configured 1-thiosugars.

Megjegyzések:Oligosaccharides and glycoconjugates are abundant in all living organisms, taking part in a multitude of biological processes. The application of natural O-glycosides in biological studies and drug development is limited by their sensitivity to enzymatic hydrolysis. This issue made it necessary to design hydrolytically stable carbohydrate mimetics, where sulfur, carbon, or longer interglycosidic connections comprising two or three atoms replace the glycosidic oxygen. However, the formation of the interglycosidic linkages between the sugar residues in high diastereoslectivity poses a major challenge. Here, we report on stereoselective synthesis of carbon-sulfur-bridged disaccharide mimetics by the free radical addition of carbohydrate thiols onto the exo-cyclic double bond of unsaturated sugars. A systematic study on UV-light initiated radical mediated hydrothiolation reactions of enoses bearing an exocyclic double bond at C1, C2, C3, C4, C5, and C6 positions of the pyranosyl ring with various sugar thiols was performed. The effect of temperature and structural variations of the alkenes and thiols on the efficacy and stereoselectivity of the reactions was systematically studied and optimized. The reactions proceeded with high efficacy and, in most cases, with complete diastereoselectivity producing a broad array of disaccharide mimetics coupling through an equatorially oriented methylensulfide bridge.

Megjegyzések:The photoinitiated thiol-ene coupling reactions of 2-substituted glycals were studied as a generally applicable strategy for stereoselective 1,2-cis-?-thioconjugation. Although all glycals reacted with full ?-selectivity, the efficacy of the reactions varied in a broad range depending on their configuration and glycals bearing axial acetoxy substituents reacted with very low efficacy at room temperature. The study revealed that the reaction progress could be promoted by cooling and inhibited by heating. At ?80 ?C, the equilibrium of the rapidly reversible addition of the thiyl radical to alkenes is shifted almost completely toward products, leading to efficient addition reactions. By exploiting this unique temperature effect a series of ?-thio-l-fucosides, -d-galactosides, and d-GlcNAc derivatives were prepared with high efficacy and complete stereoselectivity.

Megjegyzések:The applicability of the Horner-Wadsworth-Emmons olefination to the introduction of the sulfonatomethyl moiety at the 2- and 3-positions of orthogonally protectedO- and S-glycosides has been studied. The conformational preferencesand relative energies of the exo- and endocyclic alkenesulfonic acids obtained were analysed by high-temperature molecular dynamics and DFT calculations. Thioglycosides bearing a sulf-onatomethyl moiety at the secondary position have been pre-pared for the first time. Finally, the attempted synthesis of 2-sulfonatomethyl glucoside by nucleophilic substitution reactionis also described.

Megjegyzések:Staphylococcus aureus and Staphylococcus epidermidis are prominent bacterial pathogens of nosocomial infections. Both microorganisms colonize medical devices by forming adherent biofilms. Poly-[beta]-D-(1-6)-N-acetyl-glucosamine (PNAG) is a surface polysaccharide antigen which was found on both S. aureus and S. epidermidis. Animal studies have proved that PNAG can elicit antibodies which protect against staphylococcal infections. We have presented the synthesis of di-, tetra- and hexasaccharide fragments of PNAG with formyl-heptyl aglycone and their attachment to bovine serum albumin (BSA) by reductive amination.

Megjegyzések:Synthesis of carbohydrate mimetics for potential pharmaceutical applications is one of our main research topics in the Department of Pharmaceutical Chemistry at the University of Debrecen. As therapeutical application of glycosides is hampered by their in vivo instability and their low binding affinity towards proteins, we focus on the design and synthesis of unnatural derivatives with an increased stability and with better binding properties.In this article, we summarized our recent results in the following topics: i) synthesis and biological evaluation of heparinoid pentasaccharide sulfonic acids; ii) preparation of thio-linked glycosides by photoinduced free radical hydrothiolation of endocyclic double bond of sugars and application of the thiol addition method in the synthesis of self-assembling multivalent thiomannobioside ligands of mannose-binding lectins; iii) synthesis of a cysteinyl uridine pentapeptide, the first member of a novel type of peptide nucleic acids.

Megjegyzések:Idraparinux, the fully O-sulfated, O-methylated, heparin-related pentasaccharide possessing selectivefactor Xa inhibitory activity, was prepared by a new synthetic pathway. This route was based on a [2+3]block synthesis utilizing a 6-O-silyl-protected L-idose-containing trisaccharide acceptor, which wasglycosylated with a disaccharide donor containing a non-oxidized precursor of the glucuronic acid. Theunique strategy of multiple functionalizations at pentasaccharide levels, involving triple methylationfollowed by oxidation of the glucose and the idose precursors into D-glucuronic and L-iduronic acids inone step, proved to be highly efficient, providing the target pentasaccharide through a 39-step synthesisstarting from D-glucose and methyl a-D-glucopyranoside.

Megjegyzések:Introduction of a sulfonatomethyl moiety into the primary position of thioglycosides by nucleophilic displacement of the corresponding 6-O-triflate is described. The 1->6 migration of the anomeric group, which inevitably occurs through a bicyclic sulfonium ion intermediate, from conformationally flexible [beta]-thioglycosides was prevented by using an [alpha]-thioglycoside or conformationally locked [beta]-thioglycoside as the starting material. The thioglycoside 6-sulfonic acids showed excellent [alpha]-selectivity during synthesis of uronic acid containing heparinoid trisaccharides.

Megjegyzések:Syntheses of acyl protected exo-mannal derivatives were developed starting from O-peracylated mannopyranoses via the corresponding anhydro-aldose tosylhydrazones under modified Bamford-Stevens conditions. The synthesis of analogous O-peralkylated (benzylated and isopropylenated) derivatives was carried out from pyranoid and furanoid mannonolactones using methylene transfer reagents. Photoinitiated thiol?ene additions of these exo-mannals resulted in the corresponding C-(mannopyranosyl/mannofuranosyl)methyl sulfides in medium to good yields with exclusive regio- and ?(D) stereoselectivities.

Megjegyzések:A comprehensive optimization and mechanistic study on the photoinduced hydrothiolation of different d? and l? hexo? and pentoglycals with various thiols was performed, at the temperature range of RT to ?120?°C. Addition of thiols onto 2?substituted hexoglycals proceeded with complete 1,2?cis?α?stereoselectivity in all cases. Hydrothiolation of 2?substituted pentoglycals resulted in mixtures of 1,2?cis?α? and ?β?thioglycosides of varying ratio depending on the configuration of the reactants. Hydrothiolation of unsubstituted glycals at ?80?°C proceeded with excellent yields and, except for galactal, provided the axially C2?S?linked isomers with high selectivity. Cooling was always beneficial to the efficacy, increased the yields and in most cases significantly raised the stereoselectivity. The suggested mechanism explains the different conformational preferences of the intermediate carbon?centered radicals, which is a crucial factor in the stereoselectivity of the reactions.

Megjegyzések:Heparin-based anticoagulants are drugs of choice in the therapy and prophylaxis of thromboembolic diseases. Idraparinux is a synthetic anticoagulant pentasaccharide based on the heparin antithrombin-binding domain. In the frame of our ongoing research aimed at the synthesis of sulfonic acid-containing heparinoid anticoagulants,we elaborated a modular pathway to obtain a series of idraparinux-analogue pentasaccharides bearing one or two primary sulfonic acid moieties.Five protected pentasaccharides with different C-sulfonation patternswere prepared by two subsequent glycosylation reactions, respectively, using two monosaccharide and four disaccharide building blocks. Transformation of the protected derivatives into the fully O-sulfated, O-methylated sulfonic acid end-products was also studied
