Megjegyzések:The NMR pulse sequence design strategy of NORD (NO Relaxation Delay) is extended to design of two new three-module experiments, NORD {HMBC}-{HSQC-TOCSY}-{TOCSY} and NORD {HMBC}-{2BOB}-{TOCSY}, each delivering four spectra - HMBC, HSQC, TOCSY, and either HSQC-TOCSY or H2BC. Compared to individual recording of these spectra particularly the sensitivity of the least sensitive mod-ule, HMBC, is enhanced by designing the homonuclear TOCSY module to allow buildup of magnetization pertinent to HMBC during its execution. Effectively, the sensitivity of the heteronuclear modules is boosted at the expense of the inherently much higher TOCSY sensitivity, thus resulting in a significant saving in spectrometer time.

Megjegyzések:The sensitivity of cryoprobes, which are rapidly becoming available, have brought about the possibility of measurement of C-13, C-13 coupling constants at the natural abundance of C-13 using tens rather than hundreds of milligrams of compounds. This relatively recent development lays the foundation for a more routine use of the C-13, C-13 long-range coupling constants in the conformational analysis of molecules. We have designed novel H-1-detected INADEQUATE experiments optimized for long-range C-13, C-13 correlations and the measurement of long-range coupling constants. These experiments incorporate refocusing Of (1)J(CH) coupling constants prior to the formation of DQ coherences and H-1-decoupling during the long carbon-carbon evolution intervals. Such modifications significantly enhance their performance over H-1-detected INADEQUATE experiments currently in use for mapping the one-bond C-13, C-13 correlations. H-1 or C-13 polarization is used a starting point in long-range correlation H-1-detected IPAP DEPT-INADEQUATE or RINEPT-INADEQUATE experiments. These correlation experiments were modified yielding in-phase (IP) or antiphase (AP) C-13, C-13 doublets in F-1. Procedures were developed for their editing yielding accurate values of small C-13, C-13 coupling constants. The methods are illustrated using mono- and disaccharide samples and compared with related 13 C-detected experiments by means of the measurement of interglycosidic C-13, C-13 coupling constants of a disaccharide.

Megjegyzések:Here, we present a modified CPMG-HSQMBC experiment which is capable to reduce the detrimental phase twists in the ♭♭long-range'' connectivity multiplets caused by proton?proton couplings. We demonstrate that concerted CPMG pulse trains applied on both nuclei in the starting CPMG-INEPT transfer step can considerably be improved by composite p pulses that compensate for pulse imperfections and off-resonance effects. Experimental optimization of the interpulse delay within the CPMG cycle was found to be crucial in order to achieve the best possible ♭♭decoupling'' of homonuclear coupling modulation.

Megjegyzések:One-bond heteronuclear and two-bond homonuclear residual dipolar couplings measured at methylene or amine sites can be utilized as long-range constraints in structure determination of molecules as well as to facilitate characterization of local conformation by stereospecific assignment of diastereotopic protons. We present two J-modulated HMQC type experiments to measure the one-bond heteronuclear dipolar coupling contributions of geminal protons individually. In addition two-bond homonuclear residual dipolar couplings between the diastereotopic protons are also obtained.

Megjegyzések:Since the introduction of RDCs in high-resolution NMR studies of macromolecules, there is a growing interest in the development of accurate, and sensitive methods for determining coupling constants. Most methods for extracting these couplings are based on the measurement of the splitting between multiplet components in J-coupled spectra. However, these methods are often unreliable since undesired multiple-bond couplings can considerably broaden the multiplet components and consequently make accurate determination of their position difficult. To demonstrate one approach to this problem, G-BIRD(r) decoupled TROSY sequences are proposed for the measurement of 1JNH and 1JNC0 coupling constants. Resolved or unresolved splittings due to remote protons are removed by a G-BIRD(r) module employed during t1 and as a result, spectra with narrow, well-resolved peaks are obtained from which heteronuclear one-bond couplings can be accurately measured. Moreover, introduction of a spin-state-selective a/b-filter in the TROSY sequence allows the separation of the 1JNC0 doublet components into two subspectra which contain the same number of peaks as the regular TROSY spectrum. The 1JNC0 couplings are obtained from the displacement between the corresponding peaks in the subspectra.

Megjegyzések:Conventional relaxation parameters (T 1 1 , T 1 2 , and NOE), obtained at different temperatures and magnetic fields, are reported for the hydroxymethyl (C6) carbon in methyl-b-D-glucopyranoside in a D2O/DMSO cryosolvent. These data are interpreted with the Lipari?Szabo model. In addition, two-field measurements of longitudinal and spin-locked relaxation rates related to the crosscorrelated carbon?proton dipole?dipole interactions for the same carbon are reported. The complete data set consisting the conventional and cross-correlated relaxation parameters is interpreted using a new ♭♭hybrid'' approach, in which the Lipari?Szabo model for the auto-correlated spectral densities is combined with the two-site jump model for the cross-correlated spectral densities, with the global correlation time as a common parameter. The two-site jump rates thus obtained are in reasonable agreement with the ultrasonic relaxation measurements, and have reasonable temperature dependence.

Megjegyzések:A new method for the accurate determination of carbon?carbon coupling constants is described. The method is based on a modified ADEQUATE experiment, where a J-modulated spin-echo sequence precedes the ADEQUATE pulse scheme. The Jmodulation and scaling of carbon?carbon couplings is based on simultaneous incrementation of 13C chemical shift and coupling evolution periods. The time increment for the homonuclear carbon?carbon coupling evolution can be suitably scaled with respect to the corresponding increment for the chemical shift evolution. Typically a scaling factor of 2 to 3 is employed for the measurement of one-bond coupling constants, while multiplication by a factor of 10 to 15 is applied when small long-range couplings are determined. The same pulse scheme with coupling evolution period optimized for one-bond or long-range couplings allows the determination of the corresponding carbon-carbon coupling constants. The splittings of the ADEQUATE crosspeaks in the F1 dimension yield the appropriately multiplied coupling constants.
