Endomorphins-1 and -2 were substituted with all the ©a-MePhe stereoisomers in their Phe residues to generate a conformationally constrained peptide set. This series of molecules was subjected to biological assays, and for ©a-MePhe4-endomorphins-2, a conformational analysis was performed. Incorporation of (2S,3S)-©a-MePhe4 resulted in the most potent analogues of both endomorphins with enhanced enzymatic stability. Their í opioid affinities were 4-times higher than the parent peptides, they stimulated [35S]GTP₀cS binding, and they were found to be full agonists. NMR experiments revealed that C-terminal (2S,3S)-©a-MePhe in endomorphin-2 strongly favored the gauche (-) spatial orientation which implies the presence of the ?1 ) -60? rotamer of Phe4 in the binding conformer of endomorphins. Our results emphasize that the appropriate orientation of the C-terminal aromatic side chain of endomorphins is substantial for binding to the í opioid receptor.

The constitutional similarity with different secondary structure preference between the Aba-Gly and the spiro-Aba-Gly scaffolds were exploited to design the novel endomorphin-2 analogs Tyr-spiro-(R/S)-Aba-Gly-Phe-NH2 (1 and 2) and Tyr-(R/S)-Aba-Gly-Phe-NH2 (3 and 4). The (R)-spiro analog 1 was found to be a potent and selective mu-opioid agonist/partial agonist (K-i mu = 29.3 nM, IC50 = 50 nM, K-e = 0.57). NMR experiments and molecular modeling indicated that its backbone adopts mainly a U-turn in aqueous solution.
