Megjegyzések:Barleria albostellata C.B. Clarke (Acanthaceae) is a plant native to South Africa and relatively few studies have been performed on it. Species in this genus are known for their ethnopharmacological and phyto-medicinal values. In this study, the total flavonoid and phenolic contents and the antioxidant and cytotoxic activities of hexane, chloroform, and methanol extracts were evaluated at five different concentrations (15, 30, 60, 120, and 240 mu g/mL). The antioxidant activity of the extracts of B. albostellata was assessed in vitro using the 2,2 '-diphenyl-1-picrylhydrazyl (DPPH) scavenging and ferric reducing antioxidant power (FRAP) assays, while the phenolic content was determined using a Folin-Ciocalteu assay. The extracts' cytotoxicity was established using a 3-[(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide] (MTT) assay in human embryonic kidney (HEK293), cervical cancer (HeLa), and breast adenocarcinoma (MCF-7) cell lines. Methanolic leaf extracts had the highest total flavonoid content (42.39 +/- 1.14 mg GAE/g DW) compared to other solvents. Additionally, the total phenolic content was the greatest in the methanol leaf extract (6.05 +/- 0.09 GAE/g DW), followed by the methanol stem extracts (2.93 +/- 0.73 GAE/g DW). The methanolic leaf and stem extract concentrations needed for 50% inhibition (DPPH) were 16.95 mu g/mL and 14.27 mu g/mL, respectively, whereas for FRAP, the reducing powers of all extracts were considerably lower than the ascorbic acid standard. The IC50 values of extracts tested in the three cell lines were >63 mu g/mL. According to the findings of our study, the leaves and stems of B. albostellata are rich in several bioactive compounds that may be a possible source of natural antioxidants and may have the potential to treat certain diseases. The extraction of the bioactive compounds from the leaves and stems of B. albostellata using bioassay-guided fractionation and the assessment of their safety will be essential for further investigations into this species in the search of potential novel therapeutic drug leads. To the best of our knowledge, this is the first report of the cytotoxic activities of leaf and stem extracts of Barleria albostellata.

Megjegyzések:In vitro experiments were conducted to study the responses of potato (Solanum tuberosum L.) genotypes to osmotic stress. In vitro shoot cultures of 27 breeding lines and their drought-tolerant parents (referent lines: C103 and C107) were tested under osmotic stress induced by addition of PEG 6000 (Mw = 6000; 5.0, 7.5, 10.0%, w/v), D-mannitol (0.1, 0.2, 0.3 M) and PEG 600 (Mw = 600; 2.5, 5.0, 7.5%, w/v) to the Murashige-Skoog medium. Stress index (SI) was calculated from shoot length (SL) and root length (RL), root numbers (RN) and the rate of surviving shoots (SR) (SISL;RL;RN;SR = Parameter(SL;RL;RN;SR) of treated shoots/Parameter(SL;RL;RN;SR) of control shoots x 100) to compare genotypes. In the average of each breeding line and concentration, the osmotic agents resulted in SI values of 40.1, 60.8, 82.6 and 76.0 for SISL, SIRL, SIRN and SISR, respectively. In general, all SI values of C103 and SIRL,RN of C107 were significantly higher than those of the breeding lines. Nine breeding lines were found to be promising based on their final ranking. According to the results, 7.5% and 10% PEG 6000 or 0.2 M and 0.3 M D-mannitol treatments proved to be suitable for the selection of osmotic stress-tolerant genotypes.

Megjegyzések:Rooting ability of 'Royal Gala' shoots regenerated on media with different cytokinin content (thidiazurone, benzyladenine, benzyladenine-riboside and meta-topolin-riboside) was observed directly after regeneration and compared after subculture on hormone-free medium (A), or on medium with decreased cytokinin content (B), or elevated gibberellic acid (GA3) content (C) for a week, or after a subculture on proliferation media (1.0 mg l-1 benzyladenine-riboside) for four weeks (treatment D). Rooting and acclimatization of regenerants after directly regeneration was not successful. Subculture of shoots on hormone-free medium did not improve the rooting ability of shoots, while their subculture on B and C media resulted in up to 36% rooting rate depending on regeneration media from which the shoots originated. The best rooting rate (up to 76%) was achieved in shoots regenerated on medium with benzyladenine-riboside after treatment D similarly to rooting ability of micropropagated shoots (80%). The rooting capacity of shoots depended on both the cytokinin content of the regeneration media and different subculture media used. All rooted shoots survived after acclimatization.

Megjegyzések:An effective and reproducible micropropagation protocol was developed for Centratherum punctatum Cass. Successful in vitro initiation of callus and subsequent plant regeneration were obtained on nodal explants cultured on MS medium supplemented with plant growth regulators (PGRs). The maximum frequency of callus formation (98.3%) was noted on MS containing 4.0 mg/L 6-Benzylaminopurine (BAP) and 3.5 mg/L Kinetin with a maximum callus weight of 2.02 g. The best shoot induction frequency (100%) with an average of 30.2 shoots per explant was achieved when 4.5 mg/L BAP and 4.0 mg/L Kinetin were added to the MS. The same PGR combination resulted in the best callus-mediated shoot formation (8.3 shoots/callus mass). The highest rhizogenic response (95.3%) with an average 26.1 roots per shoot and root length of 6.2 cm was obtained with 1.0 mg/L Indole-3-acetic acid (IAA)-supplemented MS medium. The gas chromatography-mass spectrometry (GC-MS) technique was applied in the present study to analyze the methanolic extracts of the leaf, callus, and root of regenerated C. punctatum shoots to detect the different phytochemical constituents. The leaf extract of the regenerated C. punctatum showed 37 phytocompounds; some important bioactive compounds were the Phytol,1,6-Octadien 3,5-Dimethyl-Cis, 4,8-Dimethylnona-3,8-dien-2-one, 2,6-Octadiene, Stigmasterol, Chondrillasterol, Lanosteryl acetate, etc. In the callus, the extract had a total of 57 phytocompounds; among them, the Stigmasterol, Guanosine, and Tri-decanoic acid were the major ones. In the root extract, the GC-MS revealed a low number of 23 phytocompounds, the important compounds of which were Stigmasterol, Trimethylsilyl (TMS) derivative, Chrysantenyl 2-methuylbutanoate, 4-tert-Butoxybutan-1-ol, etc. The order in terms of numbers of phytocompounds present in tissue sources are callus > leaf > root.
