Megjegyzések:In our research, we used plants of the family Caryophyllaceae (carnations), Dianthus plumarius subsp. praecox, Dianthus giganteiformis Borbás subsp. pontederae, and Dianthus superbus L. From a conservation point of view, all three species are important in Hungary, but the most outstanding is Dianthus plumarius subsp. praecox from Aggtelek karst and the Bükk region, which has a conservation value of 100 000 HUF, while the other two species have a value of 5000 HUF. Our aim was to develop a method of gene conservation, to investigate the possibilities of in vitro propagation in ex situ programs, and to perform further morphological and analytical analyses. In plant tissue culture, the bottleneck is surface sterilisation, one of the primary steps in in vitro work, and all subsequent steps depend on the effectiveness of this procedure. In the case of protected plants, in vitro culture should be started from seed to keep gene diversity as high as possible. The propagating material was obtained from the Botanical Garden collection. Different concentrations of detergents, different exposure times, and their combinations were used to start tissue culture. The optimal disinfectant/concentration/introduction time combination was revealed during the research to produce in vitro cultures of the three carnation species with the highest values for germination, purity, and growth.

Megjegyzések:Cultivation of plants in vitro is currently one of the most common methods of vegetative propagation of plants. It makes it possible to obtain homogeneous planting material in large quantities within a short time, on small areas, and at relatively low cost; to preserve particularly valuable pheno- and genotypes, or to obtain biologically active substances of plant origin. The use of microclonal plant propagation is an excellent complement to the conservation of critical species that are endangered in their natural habitats or have difficult generative reproduction. The main goal of our research ?larification the peculiarities of tissue culture of three rare and endangered species of the Hungarian flora, viz. Dianthus plumarius ssp. praecox (Willd. ex Spreng.) Domin, Dianthus giganteiformis Borbás ssp. pontederae (Borbás) Soó, and Dianthus superbus L. Healthy and viable seeds were used as initial explants. The sterilising reagents used were different concentrations of mercuric hippochloride (HgCl2), sodium chloride (NaClO) and commercial chloramine B (NH2Cl). Subsequently, the plants were grown on Murashige and Skoog agarified hormone-free medium. It was established that for the seeds of the three researched carnation taxa, sterilization with 2,5% chloramine B for 15 minutes is the most optimal, this ensures obtaining aseptic planting material capable of intensive germination and formation of viable explants. For the induction of rhizogenesis, the most optimal is a modified MS medium with the addition of indolylacetic acid at a concentration of 0,1 mg/l.

Megjegyzések:Plants, as part of the ecological system, may find themselves in danger of various unfortunate circumstances due to their sessile nature, climate change and other human interventions/factors. Inappropriate agricultural or forestry practices, habitat destruction attributable to excessive urbanization, uncontrolled harvesting, pollution and the spread of invasive species that compete with the native ones are the main factors leading to ecological collapse. The careful protection and preservation of biodiversity is based on different conservation methods. Ex situ methods involve the protection of endangered taxa outside of their natural habitat that necessitate the location, sampling and transfer of the species in question. These are the most convenient, cost effective methods known today. In vitro plant micropropagation gives a possibility to keep alive special lines with genetic identity for conservation purposes, to multiply endangered plant species for repatriation, or producing large plant biomass for yielding secondary metabolites. In Hungary, Dianthus are classified as a protected or strictly protected species, consequently, creating a successful conservation strategy is indispensable. Therefore, different types of cytokinins (aromatic and isoprenoid side chain containing and synthetic cytokinins) applied alone or in combination were tested to determine the optimal plant growth regulator for the multiplication phase of the in vitro micropropagation of the species.

Megjegyzések:Carnation is important floral crop. The introduction of wild carnation species is carried out in parallel with research on existing species and varieties. Dianthus plumarius subsp. praecox is a wild species with an extremely high germplasm conservation importance. The most important step in the study of its cultivation is to propagate it in vitro under conditions of minimum impact on nature. Therefore, present work aimed to develop an in vitro micropropagation protocol for this species. Different types of cytokinins, such as 6-benzylaminopurine, 6-benzylaminopurine riboside, meta-topolin, N-(2-chloro-4-pyridyl)-N'-phenylurea, kinetin, N6-(2-isopentenyl) adenine, zeatin and adenine sulfate, was applied for the in vitro multiplication. Cytokinins were applied alone in a concentration of 4.5 ?M, or in the combination of 2.25 ?M BA and 2.25 ?M KIN. Their effect on the in vitro shoot multiplication and their after-effects on the in vitro rooting were tested. Based on the present detailed study, a micropropoagation protocol was developed, where high shoot multiplication rate (4.7 ? 0.4 shoot/explant with 25.1 ? 1.4-mm-long shoots) could be achieved using zeatin as cytokinin source in the shoot multiplication medium. For successful in vitro rooting 6-benzylaminopurine-containing medium was applied for 3 weeks after shoots multiplication phase on zeatin-containing medium, then shoots were conditioned for 2 weeks on plant growth regulator free medium directly before rooting. With this treatment hyperhydration of in vitro shoots could be decreased from 91 % to 12 % and high frequency (86.7 %) in vitro rooting was gained. Rooted in vitro shoots were successfully acclimatized in 71 %.
